We investigated the effect of sildenafil in protection against necrosis or apoptosis in cardiomyocytes. Adult mouse ventricular myocytes were treated with sildenafil (1 or 10 μm) for 1 h before 40 min of simulated ischemia (SI). Necrosis was determined by trypan blue exclusion and lactate dehydrogenase release following SI alone or plus 1 or 18 h of reoxygenation (RO). Apoptosis was assessed by terminal deoxynucleotidyl transferase-mediated nick end labeling assay and mitochondrial membrane…

Cyclic GMP-dependent Protein Kinase Iα Attenuates Necrosis and Apoptosis Following Ischemia/Reoxygenation in Adult Cardiomyocyte*
TLDR
Results show that opening of mitochondrial KATP channels and generation of reactive oxygen species, in association with phosphorylation of Akt, ERK, and JNK, and increased expression of NOS and Bcl-2, play an essential role in the protective effect of PKGIα.
Phospholemman Ser69 phosphorylation contributes to sildenafil-induced cardioprotection against reperfusion injury
TLDR
It is found that sildenafil enhanced Na+/K+-ATPase activity during reperfusion, providing a molecular explanation of how sildanafil guards against myocardial injury during postischemic reperfusions.
Thioredoxin-1 is required for the cardioprotecive effect of sildenafil against ischaemia/reperfusion injury and mitochondrial dysfunction in mice
TLDR
Active Trx1 is required for the onset of the cardioprotective effects of sildenafil on I/R injury, together with the preservation of cellular redox balance and mitochondrial function.
Protein Kinase G-dependent Cardioprotective Mechanism of Phosphodiesterase-5 Inhibition Involves Phosphorylation of ERK and GSK3β*
TLDR
It is demonstrated that sildenafil activates a PKG-dependent novel signaling cascade that involves activation of ERK and inhibition of glycogen synthase kinase 3β leading to cytoprotection and the first evidence for an essential role of PKG in sildanafil-induced cardioprotection is provided.
The late phase of ischemic preconditioning induces a prosurvival genetic program that results in marked attenuation of apoptosis.
PHOSPHODIESTERASE-5 INHIBITION: A NOVEL STRATEGY TO IMPROVE STEM CELL THERAPY IN THE HEART
TLDR
The data shows that in vitro inhibition of PDE-5 using either genetic or pharmacological approaches can improve stem cell therapy following myocardial infarction.
ERK phosphorylation mediates sildenafil-induced myocardial protection against ischemia-reperfusion injury in mice.
TLDR
First direct evidence is provided that PKG-dependent ERK phosphorylation is indispensable for the induction of eNOS/iNOS and Bcl-2 and the resulting cardioprotection by sildenafil.
Phosphodiesterase-5 Inhibitor, Tadalafil, Protects Against Myocardial Ischemia/Reperfusion Through Protein-Kinase G–Dependent Generation of Hydrogen Sulfide
Background— Tadalafil is a novel long-acting inhibitor of phosphodiesterase-5. Because cGMP-dependent protein kinase (PKG) signaling plays a key role in cardioprotection, we hypothesized that PKG

Sildenafil Induces Delayed Preconditioning Through Inducible Nitric Oxide Synthase–Dependent Pathway in Mouse Heart
TLDR
Data suggest that the induction of NO synthase isoforms is an essential component of the signaling mechanism for sildenafil-induced delayed preconditioning and that iNOS appears to be the primary isoform that mediates the robust cardioprotection.
Apoptosis is initiated by myocardial ischemia and executed during reperfusion.
TLDR
It is concluded that cell death in acute global ischemia followed by reperfusion occurs predominantly by necrosis and that apoptosis is of minor importance in this pathophysiological situation.
Ischemic preconditioning upregulates inducible nitric oxide synthase in cardiac myocyte.
TLDR
It is concluded that iNOS is rapidly upregulated after ischemic PC and that cardiac myocytes are the main source of isChemic PC-inducediNOS expression.
Cardioprotection with phosphodiesterase-5 inhibition--a novel preconditioning strategy.
Cardioprotection with sildenafil, a selective inhibitor of cyclic 3',5'-monophosphate-specific phosphodiesterase 5.
TLDR
It is demonstrated for the first time that within a narrow dose range, sildenafil can protect the heart from ischemia/reperfusion injury, probably through a cGMP-signaling pathway.
Sildenafil and vardenafil, types 5 and 6 phosphodiesterase inhibitors, induce caspase-dependent apoptosis of B-chronic lymphocytic leukemia cells.
TLDR
Vardenafil and sildenafil induce caspase-dependent apoptosis of B-CLL cells in vitro and thus might be considered in the treatment of CLL patients, however, further in vivo investigations should be warranted.
Essential role of inducible nitric oxide synthase in monophosphoryl lipid A-induced late cardioprotection: evidence from pharmacological inhibition and gene knockout mice.
TLDR
An obligatory role for iNOS in mediating the cardioprotective effect induced by MLA was confirmed with the pharmacological inhibition and gene knockout mice, demonstrating a direct association of infarct size reduction with increased NO production with MLA350.
Phosphodiesterase Type 5 as a Target for the Treatment of Hypoxia-Induced Pulmonary Hypertension
TLDR
PDE5 inhibition attenuates the rise in PAP and vascular remodeling when given before chronic exposure to Hypoxia and when administered as a treatment during ongoing hypoxia-induced pulmonary hypertension.
Angiotensin II Induces Apoptosis of Adult Ventricular MyocytesIn Vitro
TLDR
Ligand binding of AT1receptors on myocytes triggers PMCD by a mechanism involving PKC-mediated increases in cytosolic calcium, which result in internucleosomal DNA fragmentation.
Morphological and molecular characterization of adult cardiomyocyte apoptosis during hypoxia and reoxygenation.
TLDR
The results suggest that apoptosis predominates in cardiomyocytes after reoxygenation through a mitochondrion-dependent apoptotic pathway, and Bcl-2 prevents re oxygengenation-induced apoptosis by inhibiting cytochrome c release from the mitochondria and prevents activation of caspase-3 and -9.
