Diacerein has proved to be effective in the treatment of osteoarthritis. We investigated the effects of diacerein in animal models of carrageenin-, zymosan-, or dextran-induced paw edema and adjuvant-induced arthritis and in ovariectomized rats. In acute inflammatory models, unlike classical nonsteroidal anti-inflammatory drugs such as naproxen and ibuprofen, diacerein inhibited the rat paw edema induced by various agents. In the adjuvant-induced arthritic rats, diacerein at 100 mg/kg/day… Expand
Attenuation of inflammatory polyarthritis in TNF transgenic mice by diacerein: comparative analysis with dexamethasone, methotrexate and anti-TNF protocols
TLDR
Results indicate that diacerein has a disease modifying effect on the onset and progression of TNF driven chronic inflammatory arthritis, suggesting that the prophylactic or therapeutic potential of diACErein in patients with RA should be further examined. Expand
Study of analgesic property of diacerein in rat
TLDR
Diacerein at all the three doses possess dose dependant analgesic activity that is maximally effective up to 60 minutes and the effect persisted up to one hour only and after which it started decreasing. Expand
Diacerein protects against iodoacetate-induced osteoarthritis in the femorotibial joints of rats
TLDR
Diacerein exhibits chondroprotective effect in rats with late onset of action based on histopathological and biochemical findings, and is found to be more pronounced after 12 weeks as compared to 8 weeks in both cases. Expand
AMELIORATION OF CARRAGEENAN-INDUCED INFLAMMATION AND HYPOTHYROIDISM BY ETORICOXIB, A COX-2 INHIBITOR, IN RAT MODEL
TLDR
It is suggestive that the drug might be acting through the restoration of thyroid homeostasis which in turn ameliorates inflammation and oxidative stress. Expand
Diacerein has a weak effect on the catabolic pathway of human osteoarthritis synovial fibroblast--comparison to its effects on osteoarthritic chondrocytes.
TLDR
Diacerein does not seem to reduce but rather increase inflammatory mediators in synoviocytes, while it does not overall affect chondrocyte inflammatory profile. Expand
Curative effect of Semecarpus anacardium Linn. nut milk extract against adjuvant arthritis -- with special reference to bone metabolism.
TLDR
It can be concluded that SA possesses strong anti-arthritic property by regulating bone turnover by significantly reverted the alterations in the bone turnover observed in arthritic animals by modulating the levels of calcium, phosphorus and the activities of the enzymes names tartrate resistant acid phosphatase, acidosphatase and alkaline phosphatases. Expand
Diacerein decreases TNF-α and IL-1β levels in peritoneal fluid and prevents Baker’s yeast-induced fever in young rats
TLDR
It is suggested that diacerein partially protects against Baker’s yeast-induced fever and peritoneal leukocyte migration, and this effect appears to be due to inhibition of release of cytokines (such as TNF-α and IL-1β). Expand
Gastric antiulcer and anti-inflammatory activities of the essential oil from Casearia sylvestris Sw.
TLDR
An anti-inflammatory activity for the EOCS of Casearia sylvestris leaves is suggested beyond its anti-ulcer activity, which showed no changes related to H+ levels in the gastric secretion. Expand
The Effects of Diacerhein on Mechanical Allodynia in Inflammatory and Neuropathic Models of Nociception in Mice
TLDR
Diacerhein produces marked antiallodynic effects in carrageenan and CFA nociception models and also inhibits the neuropathic pain after PLSN, with an efficacy similar to that observed for gabapentin. Expand
Diacerein Ameliorates Liver Ischemia Reperfusion Insult in Rats
TLDR
Pretreatment with diacerein 10mg/kg improved the deteriorated liver function and the histopathological changes as well as attenuating the oxidative stress, apoptosis and inflammatory processes. Expand

Diacerein suppresses the increase in plasma nitric oxide in rat adjuvant-induced arthritis.
TLDR
The results suggest that the inhibitory effect of diacerein on rat adjUvant-induced arthritis is partly related to its reduction of the NO production induced by adjuvant- induced arthritis. Expand
Effects of diacerhein on granuloma induced cartilage breakdown in the mouse.
TLDR
The mechanism of the chondroprotective effects of diacerhein is not understood but may be explained by a reduction in the concentrations of proinflammatory cytokines. Expand
Effects of Diacerein on Indomethacin-Induced Gastric Ulceration
TLDR
The inhibitory effect of diacerein on indomethacin-induced gastric ulceration could be mediated not by the augmentation of gastric mucosal PGE2 production but by the suppression of ROS production based on its inhibition of neutrophil activation. Expand
Effect of auranofin on plasma fibronectin, C reactive protein, and albumin levels in arthritic rats.
TLDR
Auranofin, in addition to inhibiting chronic systemic paw inflammation, also altered abnormal concentrations of plasma Fn, CRP, and albumin in the adjuvant arthritic rat, thus distinguishing auran ofin from standard NSAIDs the authors have previously tested. Expand
Effects of diacerhein in an accelerated canine model of osteoarthritis.
TLDR
Diacerhein appeared to reduce the severity of fibrillation and full-thickness ulceration of the articular cartilage and the level of collagenase activity in extracts of the OA cartilage, and to increase net PG synthesis in the O a cartilage; although none of the above changes were statistically significant. Expand
Urinary hydroxy-pyridinium crosslinks provide indices of cartilage and bone involvement in arthritic diseases.
TLDR
Measurement of both pyridinium crosslinks in urine may provide information on the stage, activity, level of bone involvement and efficacy of drug therapy in arthritic diseases. Expand
Efficacy and safety of diacerein in osteoarthritis of the knee: a double-blind, placebo-controlled trial. The Diacerein Study Group.
TLDR
Diacerein, a drug for the treatment of OA, was shown to be an effective treatment for symptoms in patients with knee OA and the optimal daily dosage is 100 mg/day (50 mg twice daily). Expand
Diacerhein and rhein reduce the interleukin 1beta stimulated inducible nitric oxide synthesis level and activity while stimulating cyclooxygenase-2 synthesis in human osteoarthritic chondrocytes.
TLDR
Diacerhein and rhein, in contrast to an NSAID, are potent inhibitors of IL-1beta induced NO production by chondrocytes and cartilage, without reducing PGE2 production. Expand

TLDR
Anti-inflammatory activity was readily observed in certain steroids, pyrazolidines, salicylates and sodium aurothiomalate, and the inhibition obtained by daily treatment with the steroid paramethasone disappeared when treatment was withdrawn. Expand
Comparative study of the effect of CV-6209, a specific PAF-antagonist, on rat paw edema caused by different phlogogen agents.
TLDR
Results suggest an important role of PAF in the inflammatory process caused by intraplantar injection of different phlogogenes in the rat paw, and other drugs tested, namely indometacin, mepyramine, methysergide and nordihydroguaiaretic acid showed a more selective inhibitory profile. Expand
