Enzymatic biotransformation is the principal determinant of the pharmacokinetic properties of most antiepileptic drugs (AEDs), although some agents are excreted by the kidneys predominantly as unchanged drug. Most AEDs exhibit linear enzyme kinetics, in which changes in daily dose lead to proportional changes in serum concentration if clearance remains constant. There are several important applications of pharmacokinetics in clinical practice. Established therapeutic ranges help guide drug… Expand

Pharmacokinetic profile of levetiracetam: toward ideal characteristics.
  • P. Patsalos
  • Medicine, Biology
  • Pharmacology & therapeutics
  • 2000
TLDR
Levetiracetam has a wide margin of safety and patient-friendly pharmacokinetics that distinguish it from other currently available antiepileptic drugs, and this profile may facilitate the clinical management of patients with epilepsy by providing a safer and less-complicated therapeutic strategy. Expand
Individualizing Antiepileptic Therapy for Patients
TLDR
This chapter summarizes the lengthy experience with TDM, population pharmacokinetics (PK) modeling, and individualizing dosage regimens of older AEDs given as monotherapy and polytherapy and opens a possibility for investigation of relationship between PK and pharmacodynamic measures as well as PK/PD modeling of AED's. Expand
Therapeutic effects of ciprofloxacin on the pharmacokinetics of carbamazepine in healthy adult male volunteers.
TLDR
Results of the present study indicated that ciprofloxacin significantly increased the plasma concentration of CBZ when given concurrently to the healthy adult male volunteers. Expand
New antiepileptic drugs and preparations.
TLDR
The new preparations of phenytoin, diazepam, and valporic acid add increased efficiency in drug administration, providing a new method for prehospital treatment of seizures and a more tolerable means of administration in the ED. Expand
Advance on synthesis, pharmacokinetics, and pharmacodynamics of the anti-epileptic drug: Levetiracetam
TLDR
In this review, the synthesis, pharmacokinetics, pharmacodynamics, and reverse reaction of levetiracetam were summarized and discussed and will be helpful for the development and application ofLevetir acetam. Expand
Antiepileptic drugs—best practice guidelines for therapeutic drug monitoring: A position paper by the subcommission on therapeutic drug monitoring, ILAE Commission on Therapeutic Strategies
TLDR
Evidence from nonrandomized studies and everyday clinical experience does indicate that measuring serum concentrations of old and new generation antiepileptic drugs (AEDs) can have a valuable role in guiding patient management provided that concentrations are measured with a clear indication and are interpreted critically, taking into account the whole clinical context. Expand
Phenytoin and phenobarbital stable isotope studies in neonates.
TLDR
The use of a nonradioactive labeled isotope overcomes the limitations of conventional pharmacokinetic methodology and can be specifically useful in neonates and infants in whom volumes of distribution are rapidly changing and steady state is not achieved. Expand
The relationship between the pharmacology of antiepileptic drugs and human gene variation: An overview
TLDR
Studies of genetic variation as they relate to proteins involved in antiepileptic drug kinetics and dynamics will identify key polymorphisms in endogenous molecules that determine degrees of drug efficacy and toxicity. Expand
Anti-Arrhythmic Drugs: Pharmacokinetics and Pharmacodynamics
TLDR
This chapter will discuss the pharmacokinetics and pharmacodynamics of the commonly utilized and marketed antiarrhythmic drugs. Expand
Safety and efficacy of carbamazepine extended-release capsules in patients with bipolar disorder: QD vs BID.
  • L. Ginsberg
  • Medicine
  • Annals of clinical psychiatry : official journal of the American Academy of Clinical Psychiatrists
  • 2006
TLDR
It is suggested that CBZ-ERC dosed qd is comparable in efficacy, safety, and tolerability to CBz-ERCdosed bid for patients with bipolar disorder. Expand

Time course of carbamazepine autoinduction
TLDR
Serum concentration values obtained after 1 week of monotherapy, using the authors' dosing regimen, can be used to predict directly the dosing rate needed to obtain a specific serum concentration in most patients. Expand
Experimental studies of the influence of vigabatrin on the GABA system.
TLDR
Although no data on brain levels of the drugs are currently available, judging from vigabatrin blood concentrations in man and from information of brain levels in animals, following chronic treatment, it is conceivable that a sufficiently high concentration of the drug in human brain is obtained to augment GABA release. Expand
Test Performance Characteristics of the Serum Phenytoin Concentration (SPC): The Relationship Between SPC and Patient Response
TLDR
The retrospective evaluation of Veterans Administration Cooperative Study No. 118 suggests that the range of SPC values in successfully treated patients is quite broad; the value of the commonly accepted SPC therapeutic range in predicting various measures of patient response is quite limited; and patient response should be the ultimate end point in monitoring patients on phenytoin. Expand
Carbamazepine increases phenytoin serum concentration and reduces phenytoin clearance
TLDR
Addition of carbamazepine to pheny toin monotherapy resulted in the following significant changes: increased mean phenytoin serum concentration, decreased phenyTOin clearance, increased phenyToin elimination half-life, and increased drug-related toxicity. Expand

Duchenne muscular dystrophy
TLDR
It seems likely that, once the entire gene (cDNA) is available for screening, most DMD boys will show deletions, and the effort involved in screening all affected boys for deletions is considerably less. Expand
