Abstract Bupropion is an atypical antidepressant of the aminoketone group, structurally related to cathinone, associated with a wide interindividual variability. An extensive pharmacokinetic and pharmacodynamic review of bupropion was performed, also focusing on chemical, pharmacological, toxicological, clinical and forensic aspects of this drug without a limiting period. Bupropion is a chiral, basic, highly lipophilic drug, clinically used as racemate that undergoes extensive stereoselective…
Serotonin toxicity from isolated bupropion overdoses
TLDR
The incidence of bupropion induced serotonin toxicity is higher than reported and clinicians should monitor for serotonergic toxicity when evaluating patients after bupropions overdose.
Physiologically Based Pharmacokinetic Modeling of Bupropion and Its Metabolites in a CYP2B6 Drug-Drug-Gene Interaction Network
TLDR
A whole-body physiologically based pharmacokinetic (PBPK) model of bupropion including its DDI-relevant metabolites, and to qualify the model using clinical drug–gene interaction (DGI) and DDI data is built.
Clinical uses of Bupropion in patients with Parkinson’s disease and comorbid depressive or neuropsychiatric symptoms: a scoping review
TLDR
The available literature lacks good evidence to support the use of Bupropion in PD patients presenting depressive symptoms, and further investigations are needed to extend and confirm reported findings and to produce accurate clinical guidelines.
An updated review on synthetic cathinones.
TLDR
The present work provides a comprehensive review on history and legal status, chemistry, pharmacokinetics, pharmacodynamics, adverse effects and lethality in humans, as well as on the current knowledge of the neurotoxic mechanisms of synthetic cathinones.
Adverse effects associated with bupropion therapeutic errors in adults reported to four United States Poison Centers
TLDR
Outcomes from single unintentional ingestions of bupropion in adults are overall mild and appear to be dose related.
CLINICAL AND FORENSIC ASPECTS OF PHARMACOBEZOARS.
BACKGROUND Pharmacobezoars are specific types of bezoars formed when medicines, such as tablets, suspensions, and/or drug delivery systems, aggregate and may cause death by occluding airways with
Serotonin Syndrome in a Patient Using Paroxetine and Bupropion

Bupropion: pharmacology and therapeutic applications
TLDR
Anecdotal reports indicate bupropion may lower inflammatory mediators such as tumor necrosis factor-α, may lower fatigue in cancer and may help reduce concentration problems.
Review of bupropion.
TLDR
Bupropion is an effective antidepressant with a good side-effect profile; it is a useful alternative for patients unresponsive to or intolerant of therapeutic doses of TCAs.
Resolution of enantiomers of bupropion and its metabolites by liquid chromatography.
TLDR
This is the first attempt to review the methods of enantiosepartion of bupropion using both direct and indirect approaches in both HPLC and TLC and focuses on the separation mechanism for enantioresolution using the various methods described herein.
Metabolomics of methadone: clinical and forensic toxicological implications and variability of dose response
TLDR
Knowing the metabolomics of methadone may provide further insights, attempting a personalized therapy aiming to attain effective blood concentrations, as well as investigating clinical and forensic cases related to Methadone administration.
Antidepressant profile of bupropion and three metabolites in mice.
TLDR
Bupropion has a pharmacological spectrum in various animal models which predicts both antidepressant and stimulatory activity in man, and data suggest that the clinical antidepressant profile observed in man after administration of buPropion is related to metabolite BW 306 and possibly to BW 494, rather than to bupropion itself.
Stereoselective Metabolism of Bupropion to OH-bupropion, Threohydrobupropion, Erythrohydrobupropion, and 4′-OH-bupropion in vitro
TLDR
The stereoselective pharmacokinetics and the effect of drug-drug interactions (DDIs) and CYP2C19 pharmacogenetics on bupropion exposure were quantitatively explained by the in vitro metabolic clearances and in vivo interconversion between buPropion stereoisomers.
Metabolic profile of oxazepam and related benzodiazepines: clinical and forensic aspects
TLDR
Knowing the metabolism of oxazepam and related benzodiazepines may lead to the development of new analytical strategies for its early detection and help in further toxicological and clinical interpretations.
Chiral Plasma Pharmacokinetics and Urinary Excretion of Bupropion and Metabolites in Healthy Volunteers
TLDR
The prediction of steady-state pharmacokinetics demonstrated differential stereospecific accumulation and elimination of bupropion effect and DDI risk with CYP2D6 at steady state, which may explain observed stereoselective differences.
Identification and Structural Characterization of Three New Metabolites of Bupropion in Humans.
TLDR
The aim of this study was to determine the structures of the uncharacterized bupropion metabolites using human clinical samples and in vitro incubations, and three new metabolites were detected in human liver microsome incubations and were isolated from human urine.
Comparative metabolism of tramadol and tapentadol: a toxicological perspective
TLDR
A thorough knowledge about tramadol and tapentadol metabolomics is expected to provide additional insights to better understand the interindividual variability in their pharmacokinetics and dose-responsiveness, and contribute to the establishment of personalized therapeutic approaches, minimizing side effects and optimizing analgesic efficacy.
