Abstract The distribution of replicating DNA in the cytoplasm of somatic cells of Pteridium aquilinum was investigated, using light and electron microscope autoradiography following incorporation of 3 H-thymidine. Enzyme studies showed that over 90 % of the incorporated label occurred in DNA. In contrast to mature egg cells, at least 90 % of the labelled DNA was located in plastids and mitochondria—with very little, if any, extraorganellar DNA.
