Phosphatidylinositol 3‐kinase (PI3K) signaling controls survival and proliferation of cancer cells and is activated in around 60% of pancreatic ductal adenocarcinomas (PDACs). Although not entirely clarified, PI3K signaling is linked to cell cycle progression of PDAC cells. In this study we demonstrate that PI3K signaling controls transcription of the E2F1 gene and show that E2F1 is essential for S‐phase progression of PDAC cells. On the molecular level, PI3K signaling controls c‐myc protein…

TLDR
It is demonstrated that ATO inhibits APL cell growth through reduced expression of E2F1, cyclin E, and stimulation of pRb, and this novel mode of action of ATO in APL cells may be useful for designing new APL drugs.

TLDR
The results strongly support that VMP1-mediated autophagy may integrate the complex network of events involved in pancreatic ductal adenocarcinoma chemo-resistance and point at E2F1 and V MP1 as novel potential therapeutic targets in precise treatment strategies for pancreatic cancer.
Pancreatic Ductal Adenocarcinoma and Transcription Factors: Role of c-Myc
TLDR
Evidences showing that c-Myc function is highly dose and cell context dependent, together with its recently demonstrated ability to reprogram somatic cells towards a pluripotent stem cell-like state, indicate that the role of c- myc in pancreas pathophysiology might have been previously underscored.
DUSP1 Is a Novel Target for Enhancing Pancreatic Cancer Cell Sensitivity to Gemcitabine
TLDR
Results suggest that gemcitabine-mediated upregulation of D USP1 contributes to a negative feedback loop that attenuates its beneficial actions on stress pathways and apoptosis, raising the possibility that targeting DUSP1 in PDAC may have the advantage of enhancing gem citabine chemosensitivity while suppressing angiogenesis.
Efemp1 and p27Kip1 modulate responsiveness of pancreatic cancer cells towards a dual PI3K/mTOR inhibitor in preclinical models
TLDR
The Efemp1-p27Kip1 axis is defined as a potential marker module of PDAC cell sensitivity towards dual PI3K-mTOR inhibitors, which might help to better stratify patients in clinical trials.
Tetrandrine inhibits deregulated cell cycle in pancreatic cancer cells: Differential regulation of p21Cip1/Waf1, p27Kip1 and cyclin D1.
TLDR
It is shown for the first time that tetrandrine (TET) inhibits proliferation of the PaCa cells and inhibits PaCa tumor growth and suggests that TET may serve as a novel agent for treatment of PaCa, for which there is no effective cure to date.
KrasG12D induces EGFR-MYC cross signaling in murine primary pancreatic ductal epithelial cells
TLDR
Novel evidence is provided that EGFR signaling is needed to activate the oncogenic and pro-proliferative transcription factor c-MYC and thereby explain the crucial role of EGFR for KrasG12D-driven carcinogenesis in the pancreas.
Phosphoinositide 3-Kinase Signaling Pathway in Pancreatic Ductal Adenocarcinoma Progression, Pathogenesis, and Therapeutics
TLDR
The high incidence of mutations in thePI3K signaling cascade, accompanied by activation of parallel signaling pathways, makes PI3K a promising candidate for drug therapy and potential therapeutic opportunities for targeting pancreatic ductal adenocarcinoma are discussed.
IKKα controls canonical TGFβ–SMAD signaling to regulate genes expressing SNAIL and SLUG during EMT in Panc1 cells
TLDR
It is reported that the catalytic IκB kinase (IKK) subunit IKKα is necessary for the silencing of E-cadherin in a Panc1 cell model of TGFβ–SMAD-mediated EMT, independently of NFκB.
Critical role of hnRNP A1 in activating KRAS transcription in pancreatic cancer cells: A molecular mechanism involving G4 DNA.
TLDR
Recent advances on how KRAS is regulated in pancreatic cancer cells are illustrated, focusing on the formation of G4 structures in the KRAS promoter and their interaction with hnRNP A1.

Phosphoinositide-3-kinase signaling controls S-phase kinase-associated protein 2 transcription via E2F1 in pancreatic ductal adenocarcinoma cells.
TLDR
It is shown that PI3K controls the proliferation and G(1) phase progression of PDAC cells and that the F-box protein SKP2, an oncogene up-regulated in PDAC, is transcriptionally regulated by thePI3K/AKT1 pathway inPDAC cells.

It is demonstrated that ectopic activation of NFATc1 and the Ca2+/calcineurin signaling pathway is an important mechanism of oncogenic c‐myc activation in pancreatic cancer.

A novel IKKα‐regulated growth pathway involving the p52/RelB‐dependent transcriptional regulation of the skp2 gene is defined involving the downregulation of the F‐box protein S‐phase kinase‐associated protein 2 (skp2).
The PI 3-kinase/Akt signaling pathway is activated due to aberrant Pten expression and targets transcription factors NF-κB and c-Myc in pancreatic cancer cells

The data indicate that the aberrant expression of MMAC/PTEN contributes to the activation of the PI 3-kinase/Akt pathway and its transcription factor mediators in pancreatic cancer.
E2F1 inhibits c-Myc-driven apoptosis via PIK3CA/Akt/mTOR and COX-2 in a mouse model of human liver cancer.
TLDR
E2F1 may function as a critical antiapoptotic factor both in human and in rodent liver cancer through its ability to counteract c-Myc-driven apoptosis via activation of PIK3CA/Akt/mTOR and c- myb/COX-2 pathways.

TLDR
It is shown that GSK-3 isoforms are differentially required to maintain basal NF-kappaB DNA binding activity, transcriptional activity, and cell proliferation in Panc-1 and MiaPaCa-2 cells and that IkappaB kinase (IKK) subunits are not equally required to regulate pancreatic cancer-associated NF- kappaB activity and cell growth.
