}The objective of this work was to test the hypothesis that the limitation of nitric oxide (NO) availability accentuates microvascular reactivity to oxygen. The awake hamster chamber window model was rendered hypoxic and hyperoxic by ventilation with 10 and 100% oxygen. Systemic and microvascular parameters were determined in the two conditions and compared with normoxia in a group receiving the NO scavenger nitronyl nitroxide and a control group receiving only the vehicle (saline). Perivascular NO reduction is similar for all Hb solutions because NO binding affinities are similar; however, effects on vascular resistance are related to the type of molecular modification, molecular volume, and oxygen affinity.
pO(2) and ROS/RNS measurements in the microcirculation in hypoxia.
- MedicineMethods in molecular biology
- 2010
The real time pO(2) measurements in vivo in the microvessels and tissues of the hamster cheek pouch and window chamber preparations during normoxia and hypoxia are discussed, as well as the corresponding changes in ROS/RNS in systemic blood during normxia and Hypoxia under conditions where NO availability is maximally reduced.
Hyperoxia does not directly affect vascular tone in isolated arteries from mice
- Medicine, BiologyPloS one
- 2017
It is concluded that arteries and arterioles from healthy mice are not intrinsically sensitive to hyperoxic conditions and the present ex-vivo model is therefore not suitable for further research into mechanisms of hyperoxic vasoconstriction.

The therapeutic value of oxygen needs to be separately evaluated for late hemorrhage, as the initial blood pressure response to oxygen is the same, but it gradually decreases as the animal develops a lactic acidosis.

It is suggested that NO plays an important role not only as a regulator of peripheral vascular tone but also as a modulator of tissue oxygenation by reducing oxygen consumption by vessel walls by inhibiting NO synthesis.

Treatment with verapamil, which increases oxygen supply through vasodilatation, may further improve tissue oxygenation by lowering oxygen consumption of the microcirculation, which in this model appears to constitute an important oxygen-consuming compartment.

It is confirmed that nitrite reduction at reduced oxygen pressures is a major source for red cell NO and the formation and potential release from the red cell of this NO could have a major impact in regulating the flow of blood through the microcirculation.

The congruence of systemic and microvascular hemodynamics events found with hyperoxia suggests that the microv vascular findings are common to most tissues in the organism, and that hyperoxIA, due to vasoconstriction and the decrease of FCD, causes a maldistribution of perfusion in the microcirculation.
