Nitric oxide, superoxide, and peroxynitrite: the good, the bad, and ugly.
@article{Beckman1996NitricOS, title={Nitric oxide, superoxide, and peroxynitrite: the good, the bad, and ugly.}, author={Joseph S. Beckman and Willem H. Koppenol}, journal={The American journal of physiology}, year={1996}, volume={271 5 Pt 1}, pages={ C1424-37 } }
Nitric oxide contrasts with most intercellular messengers because it diffuses rapidly and isotropically through most tissues with little reaction but cannot be transported through the vasculature due to rapid destruction by oxyhemoglobin. The rapid diffusion of nitric oxide between cells allows it to locally integrate the responses of blood vessels to turbulence, modulate synaptic plasticity in neurons, and control the oscillatory behavior of neuronal networks. Nitric oxide is not necessarily…

It is concluded that superoxide is the limiting factor for peroxynitrite formation, which depends strongly on the availability of cellular antioxidants, and among the substances under investigation, glutathione and the synthetic compound ebselen had a major effect on preventing peroxysitic formation, whereas ascorbate failed to decrease peroxlynitrite levels.

Current evidence indicates that most of the cytotoxicity attributed to NO is rather due to peroxynitrite, produced from the diffusion-controlled reaction between NO and another free radical, the superoxide anion, which is presented in detail in this review.

Evidence is presented of the first evidence that peroxynitrite (ONOO(-)) releases zinc from the zinc-thiolate cluster of endothelial NOS (eNOS) and presumably forms disulfide bonds between the monomers, providing a novel mechanism for modulation of the enzyme function in disease.
