In order to enhance the yield and productivity of metabolite production, researchers have focused almost exclusively on enzyme amplification or other modifications of the product pathway. However, overproduction of many metabolites requires significant redirection of flux distributions in the primary metabolism, which may not readily occur following product deregulation because metabolic pathways have evolved to exhibit control architectures that resist flux alterations at branch points. This…

Engineering Cellular Metabolism
Pathway analysis, engineering, and physiological considerations for redirecting central metabolism.
TLDR
The results suggest that some glycolytic intermediates may serve as a signal in the regulation of the phosphotransferase system, heat shock response, and nitrogen regulation, however, the role of central metabolites in these regulations has not been determined conclusively.
Metabolic Flux Analysis
TLDR
Metabolic engineering emerged then as the scientific field aiming at the directed modification of the enzymatic, regulatory, or transport activities of the cell to improve cellular properties, with the use of recombinant DNA technology.
Quantifying and directing metabolite flux: Application to amino acid overproduction
TLDR
By applying molecular techniques, to selected targets of the l-lysine and l-isoleucine biosynthesis of Corynebacterium glutamicum, high amino acid overproduction can be obtained with this bacterium.

Metabolic Pathway Rates and Culture Fluorescence in Batch Fermentations of Clostridium Acetobutylicum
TLDR
Stoichiometric relationships in the metabolic pathway of Clostridium acetobutylicum are used to formulate a framework for the analysis of pathway rates and intracellular NAD(P)H fluorescence measurements are applied to two batch fermentations.
Identification of metabolic model: Citrate production from glucose by Candida lipolytica
TLDR
The concept of mass balance was used to analyze the metabolic pathways of citrate production by Candida lipolytica from glucose, and models I, II, and III were found to be most unlikely because of the unusual flux assessed from these models.

TLDR
Data in vitro and in vivo lead to the conclusion that the activation of prephenate dehydratase by leucine or methionine is primarily significant under conditions of low phenylalanine (inhibitor) concentration.
