The FKHR gene was first identified from its disruption by the t(2;13) chromosomal translocation seen in the pediatric tumor alveolar rhabdomyosarcoma. It encodes for a member of the forkhead family of transcription factors. Recently, a homolog of FKHR in the nematode Caenorhabditis elegans was identified called DAF-16, which is a downstream target of two Akt homologs in an insulin-related signaling pathway. We have examined the possible role of Akt in the regulation of FKHR. We find that FKHR…

Forkhead Family Transcription Factor FKHRL1 Is Expressed in Human Megakaryocytes
TLDR
The results suggest that FKHRL1 plays an important role in the cell cycle of megakaryocytic cells as one of the downstream target molecules of phosphatidylinositol 3-kinase-Akt, presumably mediated through the activation or inactivation of cell cycle-associated gene(s).
Roles of the forkhead in rhabdomyosarcoma (FKHR) phosphorylation sites in regulating 14-3-3 binding, transactivation and nuclear targetting.
TLDR
It is found that the overexpression of FKHR[S256A] (where Ser-256-->Ala) blocks PKB activity in cells, preventing phosphorylation of the endogenous substrates FKHRL1 and glycogen synthase kinase-3.
The Forkhead Transcription Factor AFX Activates Apoptosis by Induction of the BCL-6 Transcriptional Repressor*
TLDR
An AFX-regulated pathway that appears to account for at least part of this apoptotic regulatory system is described, suggesting that AFX regulates apoptosis in part by suppressing the levels of anti-apoptotic BCL-XL through the transcriptional repressor BCL6.
FOXO Transcription Factors as Regulators of Immune Homeostasis: Molecules to Die for?
TLDR
A greater understanding of the regulation and function of this signaling pathway in lymphocytes may provide novel therapeutic opportunities for immune diseases.
The kinase DYRK1A phosphorylates the transcription factor FKHR at Ser329 in vitro, a novel in vivo phosphorylation site.
TLDR
It is found that FKHR and DYRK1A co-localize in discrete regions of the nucleus and can be co-immunoprecipitated from cell extracts, which suggest that DYRk1A may phosphorylate FK HR at Ser(329) in vivo.
Transforming growth factor beta enhances epithelial cell survival via Akt-dependent regulation of FKHRL1.
TLDR
TGF beta may play a role in epithelial cell survival via Akt-dependent regulation of FKHRL1 via dominant-negative Akt, which abrogated the antiapoptotic effect of TGF beta.
Regulation of nuclear translocation of forkhead transcription factor AFX by protein kinase B.
TLDR
Results indicate that the nuclear translocation of AFX is negatively regulated through its phosphorylation by PKB, a downstream target of PI 3-kinase.
Transcriptional regulation of neuronal genes and its effect on neural functions: expression and function of forkhead transcription factors in neurons.
TLDR
Sodium orthovanadate, a protein tyrosine phosphatase inhibitor, up-regulated Akt activity in the brain and in turn rescue neurons from delayed neuronal death by inhibiting FKHR-dependent or -independent death signals in neurons.
Characterization of Insulin Inhibition of Transactivation by a C-terminal Fragment of the Forkhead Transcription Factor Foxo1 in Rat Hepatoma Cells*
TLDR
It is concluded that the transcriptional activity of Foxo1 is regulated at different levels by insulin: transactivation, as well as DNA binding and nuclear exclusion, allow the precise control of transcription ofFoxo1 target genes by insulin.

The alveolar rhabdomyosarcoma PAX3/FKHR fusion protein is a transcriptional activator.
TLDR
The ability of the PAX3/FKHR hybrid protein to bind DNA in a sequence specific manner and to transactivate the expression of artificial reporter genes suggests that its aberrant expression could subvert the transcriptional programs that normally control the growth, differentiation, and survival of primitive myogenic precursors in vivo.
daf-16: An HNF-3/forkhead family member that can function to double the life-span of Caenorhabditis elegans.
The wild-type Caenorhabditis elegans nematode ages rapidly, undergoing development, senescence, and death in less than 3 weeks. In contrast, mutants with reduced activity of the gene daf-2, a homolog
Identification of nine tissue-specific transcription factors of the hepatocyte nuclear factor 3/forkhead DNA-binding-domain family.
TLDR
The isolation of nine additional HNF-3/fkh homologue (HFH) clones from rodent tissue cDNAs by using both low-stringency hybridization and a polymerase chain reaction protocol is described.
The Fork head transcription factor DAF-16 transduces insulin-like metabolic and longevity signals in C. elegans
TLDR
It is shown that null mutations in Daf-16 suppress the effects of mutations in daf-2 or age-1; lack of dAF-16 bypasses the need for this insulin receptor-like signalling pathway.
Fusion of a fork head domain gene to PAX3 in the solid tumour alveolar rhabdomyosarcoma
TLDR
Findings indicate that the t(2;13) generates a potentially tumorigenic fusion transcription factor consisting of intact PAX3 DNA binding domains, a truncated fork head DNA binding domain and C–terminal FKHR regions.
Role of Translocation in the Activation and Function of Protein Kinase B*
TLDR
The results indicate that PI3-K activity is required for translocation of PKB to the plasma membrane, where its activation occurs through phosphorylation of the same sites that are induced by insulin or IGF-1.
Hepatic nuclear factor 3- and hormone-regulated expression of the phosphoenolpyruvate carboxykinase and insulin-like growth factor-binding protein 1 genes
TLDR
A model in which insulin is postulated to mediate its negative effect on glucocorticoid-induced PEPCK and IGFBP-1 gene transcription indirectly by inhibiting HNF-3 action is proposed.
Mitogenic Activation, Phosphorylation, and Nuclear Translocation of Protein Kinase Bβ*
TLDR
Observations provide evidence that PKBβ undergoes nuclear translocation upon mitogenic activation and support a role for PKB in signaling from receptor tyrosine kinases to the nucleus through phosphatidylinositol 3-kinase.
Wild type PAX3 protein and the PAX3-FKHR fusion protein of alveolar rhabdomyosarcoma contain potent, structurally distinct transcriptional activation domains.
TLDR
To compare the PAX3 and putative PAX3-FKHR transactivation domains, deletion mapping demonstrated essential acidic and/or serine/threonine-rich domains in the extreme 3' ends of their respective coding regions and positive modifying elements in adjacent 5' sequences and suggest that a consequent difference in function is important for oncogenesis.
Inhibition of glycogen synthase kinase-3 by insulin mediated by protein kinase B
TLDR
It is shown that agents which prevent the activation of both MAPKAP kinase-1 and p70S6k by insulin in vivo do not block the phosphorylation and inhibition of GSK3, and it is demonstrated that PKB is the product of the proto-oncogene protein kinase B (PKB, also known as Akt/RAC).
