Poly(ADP-ribose) polymerases (PARPs) are enzymes that catalyze the transfer of ADP-ribose units from β-nicotinamide adenine dinucleotide (NAD+) to acceptor proteins. PARP-1 is responsible for more than 90 % of protein poly-ADP-ribosylation in the brain and may play a role as a molecular switch for cell survival and death. The functional roles of PARP-1 are largely mediated by its activation after binding to damaged DNA. Upon binding, PARP-1 activity increases rapidly and cleaves NAD+ into ADP…

The Sound of Silence: RNAi in Poly (ADP-Ribose) Research

TLDR
The potential of RNAi to manipulate the levels of PARPs and PARG, and consequently those of PAR and ADPR, are discussed, and the results with those obtained after genetic or chemical disruption are compared.

Inputs and outputs of poly(ADP-ribosyl)ation: Relevance to oxidative stress

Poly(ADP-Ribose) Polymerases in Host-Pathogen Interactions, Inflammation, and Immunity

TLDR
Roles played by PARP enzymes in host-pathogen interactions and in diseases with an associated inflammatory response are focused on, and PARP inhibitors show promise in the treatment of inflammatory disorders and conditions with an inflammatory component.

PARP inhibitors and the Kidney

TLDR
Current data on the kidney effects of PARPis is reviewed and opens the door to further research in this arena as safe dosing of these agents in patients with advanced CKD and end stage kidney disease (ESKD) is yet to be determined.

Inhibition of Poly(ADP-ribose) Polymerase-1 Enhances Gene Expression of Selected Sirtuins and APP Cleaving Enzymes in Amyloid Beta Cytotoxicity

TLDR
PARP1 inhibition, besides activation of nuclear SIRTs and mitochondrial Sirt4 expression, enhanced transcription of enzyme(s) involved in βAPP metabolism, and this effect should be considered in its application against Aβ peptide toxicity.

[THE EFFECT OF OF POLY(ADP-RIBOSE) POLYMERASE INHIBITOR 4-HYDROXY-QUINAZOLINE ON DEATH OF IMMUNE CELLS UNDER IMMUNE COMPLEX-MEDIATED INJURY IN MICE].

TLDR
The results suggest that PARP may be involved in thymic and lymph node cell damage in immune complex mediated pathology and give evidence that inhibition of this enzyme may constitute a perspective target inimmune complex diseases prevention and therapy.

Adenoviral vectors for modulation of poly(ADP-ribose) polymerase-1 (PARP1) – dependent DNA repair as a predictive tool for chemotherapy

TLDR
The set of adenoviral vectors presented in this study can be used to modulate PARP activity in normal human cells and may therefore be used as a tool for testing cellular sensitization by blocking DNA BER by administration of PARP inhibitors and to gain better understanding of potential PARP inhibitor resistance.

TLDR
It is shown that in PARP-1−/− glial cells a loss of several stress-activated transcription factors as well as decreased expression of genes for cytokines and cellular adhesion molecules is demonstrated and that augmented expression of some of these genes is independent of PARp-1 catalytic activity.

TLDR
The hypothesis that PARPs might regulate cell fate as essential modulators of death and survival transcriptional programs is discussed with relation to nuclear factor kappaB and p53.

Minocycline inhibits poly(ADP-ribose) polymerase-1 at nanomolar concentrations.

TLDR
It is reported that PARP-1 enzymatic activity is directly inhibited by minocycline and other tetracycline derivatives that have previously been shown to have neuroprotective and anti-inflammatory actions.

This review summarizes recent findings on PARP1 and puts them in the context of metabolic stress and inflammation in aging.

Poly(ADP-ribosyl)ation regulation of life and death in the nervous system.

TLDR
The emerging mechanism dictating the fate of neurons appears to involve the regulation of PAR levels in neurons, and therapies targeting poly(ADP-ribosyl)ation in the treatment of neurodegenerative conditions such as stroke and Parkinson's disease are required.

Inhibition of poly(ADP-ribose) polymerase-1 attenuates the toxicity of carbon tetrachloride

TLDR
It is demonstrated that the hepatotoxicity of CCl4 is dependent on the overactivation of PARP-1 and that inhibition of this enzyme attenuates the hepatotinamide adenine dinucleotide and adenosine triphosphate toxicity of C Cl4.
