For many years now, scientific and technological research has witnessed a dramatic increase of interest in studies of single molecules. A large number of ongoing projects are dealing with examinations of individual particles, and many of them have been funded by the Volkswagen Stiftung. Over the years, results and outlooks of these projects have been presented at biannual symposia. This year’s meeting took place from February 22–25 in Staffelstein, Germany and was organized by Stefan Hell…

CORRIGENDUM: Controlled positioning of a DNA molecule in an electrode setup based on self-assembly and microstructuring
TLDR
An approach to place single molecules at a defined position on a microstructured surface based on self-assembly steps is described and opens the way for access to the molecular world with high throughput and without the need for ultramicroscopical setups.
Parallel confocal detection of single biomolecules using diffractive optics and integrated detector units.
TLDR
This mini-review article presents recent extensions aimed to implement parallel laser excitation and parallel fluorescence detection that can lead to even further increase in throughput in miniaturised array-based analytical systems.
Single-Molecule Optomechanical Cycle
TLDR
The polymer was found to contract against an external force acting along the polymer backbone, thus delivering mechanical work, demonstrating for the first time optomechanical energy conversion in a single-molecule device.
Reversible intramolecular electron transfer at the single-molecule level.
TLDR
Evidence for intramolecular (excitation) energy migration, that is, energy hopping and directional F%rster energy transfer is shown and electron transfer between a guanine base and a dye linked to a DNA molecule is monitored.
DNA: A Programmable Force Sensor
TLDR
This work introduces an approach in which the unbinding forces required to break intermolecular bonds are measured in a differential format by comparison with a known reference bond (here, a short DNAduplex), which allows for highly specific parallel assays.
Probing the free-energy surface for protein folding with single-molecule fluorescence spectroscopy
TLDR
It is shown that single-molecule FRET measurements of a small cold-shock protein expose equilibrium collapse of the unfolded polypeptide and allow us to calculate limits on the polyPEptide reconfiguration time.
Chiral recognition in dimerization of adsorbed cysteine observed by scanning tunnelling microscopy
TLDR
Scanning tunnelling microscopy studies of cysteine adsorbed to a (110) gold surface show that molecular pairs formed from a racemic mixture of this naturally occurring amino acid are exclusively homochiral, and that their binding to the gold surface is associated with local surface restructuring.

TLDR
A family of concepts has emerged that overcomes the diffraction barrier altogether and, relying on saturated optical transitions, these concepts are limited only by the attainable saturation level.
Multistep energy transfer in single molecular photonic wires.
We demonstrate the synthesis and spectroscopic characterization of an unidirectional photonic wire based on four highly efficient fluorescence energy-transfer steps (FRET) between five spectrally
