Studies have indicated that oligodendrocytes in the spinal cord originate from a ventral progenitor domain defined by expression of the oligodendrocyte-determining bHLH proteins Olig1 and Olig2. Here, we provide evidence that progenitors in the dorsal spinal cord and hindbrain also produce oligodendrocytes and that the specification of these cells may result from a dorsal evasion of BMP signaling over time. Moreover, we show that the generation of ventral oligodendrocytes in the spinal cord… Expand
A subset of oligodendrocytes generated from radial glia in the dorsal spinal cord
TLDR
It is found that a subpopulation of oligodendrocytes is generated from the Dbx1-expressing domain of the VZ, spanning the dorsoventral midline, and formed late during embryogenesis by transformation of radial glia and settle mainly in the lateral white matter. Expand
Generation of Oligodendrocyte Precursor Cells from Mouse Dorsal Spinal Cord Independent of Nkx6 Regulation and Shh Signaling
TLDR
In this study, in vivo evidence for a late phase of Olig gene expression independent of Nkx6 and Shh gene activities is provided and a brief second wave of oligodendrogenesis in the dorsal spinal cord is revealed. Expand
Dorsally‐derived oligodendrocytes in the spinal cord contribute to axonal myelination during development and remyelination following focal demyelination
TLDR
In response to a focal demyelination injury, a large number of newly differentiated oligodendrocytes originated from d OLs, suggesting that dOLs may provide an important source of OPCs for axonal remyelinations in multiple sclerosis or spinal cord injury. Expand
FGF signaling controls Shh-dependent oligodendroglial fate specification in the ventral spinal cord
TLDR
The data reveal that the FGF signaling pathway is activated and required for OPC commitment in the ventral spinal cord, and may prove important in defining strategies to produce large populations of determined oligodendrocyte precursor cells from undetermined neural progenitors, including stem cells. Expand
Origins of oligodendrocytes in the cerebellum, whose development is controlled by the transcription factor, Sox9
TLDR
Evidence is obtained that the majority of oligodendrocytes in the cerebellum originate from the Olig2-expressing neuroepithelial domain in the ventral rhombomere 1 (r1), while about 6% of cerebellar oligodends are produced in the Cerebellar ventricular zone. Expand
Diminished ventral oligodendrocyte precursor generation results in the subsequent over-production of dorsal oligodendrocyte precursors of aberrant morphology and function
TLDR
It is shown that ablating Shh signaling from nascent ventricular zone derivatives and partially from the floor plate results in a severe diminishment of ventral derived OPCs but normal numbers of motor neurons in the postnatal spinal cord, suggesting that vOPCs have a unique ability to communicate with injured motor neurons.
Expand
Netrin-1 Is Required for the Normal Development of Spinal Cord Oligodendrocytes
TLDR
It is shown that in the spinal cord of netrin-1 mutant mice, oligodendrocyte precursors failed to disperse from the ventral midline as a consequence of a lack of polarization and directional migration and the lack of netin-1 resulted in an overall reduction of oligodendedrocytes lineage cells that was independent of the failure of initial dispersal. Expand
Multiple roles for olig2 in the hindbrain: Oligodendrocyte and abducens motor neuron specification and facial motor neuron migration
TLDR
Cranial motor neuron and OPC development in zebrafish is investigated and it is found that somatic abducens motor neurons and a small subset of OPCs arise from common olig2 neuroepithelial precursors in rhombomeres r5 and r6, but that all otherMotor neurons and O PCs do not similarly develop from shared pools of olig2 precursor. Expand
FGFs are orchestra conductors of Shh-dependent oligodendroglial fate specification in the ventral spinal cord
TLDR
It is shown that ventral spinal progenitor cells activate FGF signaling at the onset of oligodendrocyte precursor cell (OPC) generation, as do they dorsal counterpart, and that FGFs serve dual essential functions in ventral OPC specification.

Expand
Oligodendrocyte precursors originate from both the dorsal and the ventral parts of the spinal cord
TLDR
It is shown that oligodendrocytes are generated in vivo from both ventral and dorsal halves of the neural tube, and extensive ventrodorsal, as well as dorsoventral, migrations of oligodENDrogenic cells take place during spinal cord differentiation. Expand
Local control of oligodendrocyte development in isolated dorsal mouse spinal cord
TLDR
The data suggest that early embryonic dorsal mouse spinal cord has an independent potential to generate oligodendrocytes under appropriate conditions, whether this potential is realized during normal spinal cord development is currently unknown. Expand
Bone morphogenetic proteins negatively control oligodendrocyte precursor specification in the chick spinal cord.
TLDR
The data show that oligodendrogenesis is not only regulated by ventral inductive signals such as Shh, but also by dorsal inhibiting signals including BMP factors, and suggest that the dorsoventral position of OLPs depends on a tightly regulated balance between Shh and BMP activities. Expand
FGF-dependent generation of oligodendrocytes by a hedgehog-independent pathway
TLDR
It is shown that NSCs isolated from the dorsal spinal cord can generate oligodendrocytes following FGF2 treatment, a MAP kinase dependent phenomenon that is associated with induction of the obligate oligogenic gene Olig2. Expand
Dorsal Spinal Cord Neuroepithelium Generates Astrocytes but Not Oligodendrocytes
TLDR
In mixed cultures of ventral and dorsal cells, only ventral cells generated oligodendrocytes, whereas both ventraland dorsal cells generated astrocyte, therefore, oligod endocrine cells are derived specifically from ventral neuroepithelium, and astroCytes from both dorsal and ventral. Expand
Control of oligodendrocyte differentiation by the Nkx2.2 homeodomain transcription factor.
TLDR
The results strongly suggest that Nkx2.2 regulates the differentiation and/or maturation, but not the initial specification, of oligodendrocyte progenitors, and that overproduction of Nk X.2 protein in fibroblast cells can induce gene expression from the proteolipid protein promoter. Expand
Region-specific and stage-dependent regulation of Olig gene expression and oligodendrogenesis by Nkx6.1 homeodomain transcription factor
TLDR
It is reported that Nkx6.1 also controls oligodendrocyte development in the developing spinal cord, possibly by regulating Olig gene expression in the ventral neuroepithelium. Expand
Dorsal and intermediate neuronal cell types of the spinal cord are established by a BMP signaling pathway.
TLDR
It is demonstrated that Bmp signaling is essential for the establishment of the prospective neural crest and dorsal sensory Rohon-Beard neurons of the spinal cord, and a presumptive ventral signal, Hh signaling, acts to restrict the amount of dorsal sensory neurons and trunk neural crest. Expand
Regulation of oligodendrocyte development in the vertebrate CNS
TLDR
Understanding the regulation of oligodendrogenesis will facilitate the use of these endogenous precursors to enhance repair in a variety of pathological conditions. Expand
