Aspergillus niger PA2, a novel strain isolated from waste effluents of food industry, is a potential extracellular tyrosinase producer. Enzyme activity and L-DOPA production were maximum when glucose and peptone were employed as C source and nitrogen source respectively in the medium and enhanced notably when the copper was supplemented, thus depicting the significance of copper in tyrosinase activity. Tyrosinase-encoding gene from the fungus was cloned, and amplification of the tyrosinase gene…

Novel means of quantitatively expressing tyrosinase activity is described in this article, which uses a set of parameters obtained from non-linear estimation of the progress curves and is suitable for enzymatic reactions which do not comply with Michaelis–Menten kinetics.
Antimicrobial Enzymes in Milk, and Their Role in Human Milk
This chapter describes the antibacterial properties and mode of action of antimicrobial enzymes in human milk, and compares it with other species to improve the understanding of the role of these enzymes in antibacterial defence strategies in nature and provide a basis for the development and improvement of applications of these enzyme as antibacterial agents.
Anticancer component identification from the extract of Dysosma versipellis and Glycyrrhiza uralensis based on support vector regression and mean impact value
The good therapeutic effect of herbal medicines depends on their abundant components and it's extremely necessary to find out the main bioactive ingredients. In this paper, the extract of Dysosma

Cloning and characterization of a tyrosinase gene from the white-rot fungus Pycnoporus sanguineus, and overproduction of the recombinant protein in Aspergillus niger
A new tyrosinase-encoding gene and the corresponding cDNA from the white-rot fungus Pycnoporus sanguineus BRFM49 were cloned and shown to be over-expressed in Aspergillus niger, a particularly suitable fungus for heterologous expression of proteins of biotechnological interest.
Identification of copper ligands in Aspergillus oryzae tyrosinase by site-directed mutagenesis.
The most likely binding sites of tyrosinase for Cu(A) are His-63, His-84 and His-93, with the remaining conserved Cys-82 providing the fourth ligand, according to the correlation of g( parallel) and (Cu)A( parallel), which deduced that the nitrogen or sulphur donors in the copper ligands should be in a square or a distorted tetrahedral geometric environment.
Purification, Characterization, and Molecular Cloning of Tyrosinase from Pholiota nameko
The results suggest that P. nameko tyrosinase is expressed as a proenzyme followed by specific cleavage to produce a mature enzyme.
Cloning, expression and characterisation of two tyrosinase cDNAs from Agaricus bisporus
Alignment and phylogenetic analysis of 35 tyrosinase and polyphenol oxidase sequences of animal, plant, fungal, and bacterial origin indicated conserved copper-binding domains, and stronger conservation within genera than between them.

Aspergillus niger PA2: a novel strain for extracellular biotransformation of l-tyrosine into l-DOPA
Aspergillus niger PA2 could be a promising resource and may be further exploited for large-scale production of l-DOPA, the most widely used drug of choice for the treatment of Parkinson’s disease and other neurologic injuries.
Cloning and expression of a tyrosinase from Aspergillus oryzae in Yarrowia lipolytica: application in l-DOPA biotransformation
Amplification of the tyrosinase gene (melO) from the genomic DNA of Aspergillus oryzae NCIM 1212 yielded a 1.6-kb product, and the resulting pTyro-YLEX1 vector was transformed in Yarrowia lipolytica strain Po1g, indicating the splicing of an intron present in the melO gene by Po1G.
