The highly conserved spindle assembly checkpoint (SAC) ensures that the sister chromatids of the duplicated genome are not separated and distributed to the spindle poles before all chromosomes have been properly linked to the microtubules of the mitotic spindle. Biochemically, the SAC delays cell cycle progression by preventing activation of the anaphase-promoting complex (APC/C) or cyclosome; whose activation by Cdc20 is required for sister-chromatid separation, which marks the transition into…

Trends in mathematical modeling of host–pathogen interactions
TLDR
This review highlights the current trends in mathematical modeling approaches and related methods used for understanding host–pathogen interactions and discusses the phenomena of molecular mimicry and crypsis as well as the interplay between defense and counter defense.
Multi-scale stochastic organization-oriented coarse-graining exemplified on the human mitotic checkpoint
TLDR
It is concluded that understanding, modeling and analyzing complex bio-molecular systems can greatly benefit from a set of coarse-graining methods that, ideally, can be automatically applied and that allow the different levels of abstraction to be related.

TLDR
In this study, bistable mathematical models for both activation and silencing of mitotic checkpoints were constructed and analyzed and a one-parameter bifurcation was computed to show the realistic biochemical switches considering all signals.

TLDR
Qualitative models of the human SAC should account for the positive feedback on APC/C activation driven by the kinetochores which is essential for SAC silencing, and the presented analysis allows for systems-level understanding of mitotic control.
Chelidonine induces mitotic slippage and apoptotic-like death in SGC-7901 human gastric carcinoma cells.
TLDR
Chelidonine induces M phase arrest and mitotic slippage of SGC-7901 human gastric carcinoma cells via downregulating the expression of BubR1, Cdk1 and cyclin B1 proteins and apoptosis-associated protein, caspase-3.

Active Transport Can Greatly Enhance Cdc20:Mad2 Formation
TLDR
It is shown that an active transport of O-Mad2 towards the spindle mid-zone increases the efficiency of Mad2-activation and this mechanism can greatly enhance the formation of Cdc20:Mad2 and gives an explanation on how the “wait-anaphase” signal can dissolve abruptly within a short time.
Running on a treadmill: dynamic inhibition of APC/C by the spindle checkpoint
TLDR
A dynamic model for APC/CCdc20 regulation by MCC is suggested in whichAPC/C- and Mad3-dependent ubiquitination of Cdc20 constitutes a self-regulated switch that rapidly inactivates the spindle checkpoint upon correct chromosome attachment.
Structure of the mitotic checkpoint complex
TLDR
This study shows how APC/C inhibition is coupled to degron recognition by co-activators and how p31comet disrupts MCC assembly to antagonize the SAC.
Inhibitory factors associated with anaphase-promoting complex/cylosome in mitotic checkpoint
TLDR
Contrary to the original MCC hypothesis, it is found that MCC disassembles upon exit from the mitotic checkpoint, so the requirement of the mitosis checkpoint system for the binding of Mad2 and BubR1 to Cdc20 may be for the assembly of the inhibitory complex rather than for CDC20 sequestration.
Two different mitotic checkpoint inhibitors of the anaphase-promoting complex/cyclosome antagonize the action of the activator Cdc20
TLDR
The mitotic checkpoint system is studied in extracts that reproduce its downstream events and it is found that inhibitory factors are associated with APC/C in the checkpoint-arrested state, which can be recovered from immunoprecipitates.
Structure of the Anaphase-Promoting Complex/Cyclosome Interacting with a Mitotic Checkpoint Complex
TLDR
Three-dimensional models of human APC/C in various functional states revealed that MCC associates with the Cdc20 binding site on APc/C, locks the otherwise flexible APC-C in a “closed” state, and prevents binding and ubiquitylation of a wide range of different APC /C substrates.
The APC/C maintains the spindle assembly checkpoint by targeting Cdc20 for destruction
The spindle assembly checkpoint (SAC) is required to block sister chromatid separation until all chromosomes are properly attached to the mitotic apparatus. The SAC prevents cells from entering
Modeling the temporal evolution of the spindle assembly checkpoint and role of Aurora B kinase
TLDR
The current model of the spindle assembly checkpoint is robust to variation in its key diffusion-limited parameters and when Aurora B inhibition is considered within the model, for a certain range of inhibitor concentrations, a prolonged prometaphase/metaphase is observed.

TLDR
BubR1 and Mad2 act cooperatively to prevent premature separation of sister chromatids by directly inhibiting APC, and quantitatively inhibit Cdc20-APC at physiological concentrations.
