Cyclophosphamide (CYP) and methotrexate (MTX) have been evaluated for their ability to induce toxicity in human peripheral blood lymphocytes (PBLs) and the protective role of mitochondrial and lysosomal stabilizing agents. The potential toxicity effects of CYP and MTX were measured in vitro by cellular parameters assays such as cellular viability, reactive oxygen species (ROS) formation, mitochondrial membrane permeability transition (mitochondrial membrane potential (MMP)) collapse, lysosomal…

Trifluoperazine an Antipsychotic Drug and Inhibitor of Mitochondrial Permeability Transition Protects Cytarabine and Ifosfamide-Induced Neurotoxicity

Insight is provided into the mechanism of protection by TFP against Ara-C and IFOS neurotoxicity, which is related, to neuronal ROS formation and mitochondrial damages.

Ameliorative effect of herbacetin against cyclophosphamide-induced nephrotoxicity in rats via attenuation of oxidative stress, inflammation, apoptosis and mitochondrial dysfunction

Results indicate that HBN might be considered as a potential protective agent against nephrotoxicity, and may be due to its antioxidant, anti-inflammatory, and anti-apoptotic potential.

Contrasting Role of Dose Increase in Modulating Sofosbuvir-Induced Hepatocyte Toxicity

It is concluded that sofosbuvir while looking toxic and pro-oxidant in lower concentrations, acts as protective and antioxidant in higher concentrations and acts as an antioxidant and hepatoprotective.

A new approach on lithium-induced neurotoxicity using rat neuronal cortical culture: Involvement of oxidative stress and lysosomal/mitochondrial toxic Cross-Talk

It is concluded that Li causes neurotoxicity in a dose-dependent manner and is a result of the generation of ROS and LP, which leads to mitochondrial/lysosomal toxic cross-talk.

Hydroxychloroquine Attenuates Acute Inflammation (LPS)-Induced Apoptosis via Inhibiting TRPV1 Channel/ROS Signaling Pathways in Human Monocytes

The present data suggest TRPV1 activation through the acute INF (LPS)-induced inflammatory, oxidant, and apoptotic adverse actions in monocyte cells, whereas HCQ prevented adverse actions via the modulation of TRpV1.

Quercetin, chrysin, caffeic acid and ferulic acid ameliorate cyclophosphamide-induced toxicities in SH-SY5Y cells

Use of antioxidant as nutritional supplements, and in particular of caffeic acid, chrysin, quercetin and ferulic acid, reduce apoptotic effects of CP in SH-SY5Y cells that could add insight into therapeutic approaches to CP-induced cell injuries.

Risperidone Toxicity on Human Blood Lymphocytes in Nano molar Concentrations.

It could be concluded that oxidative stress is one of the mechanisms of risperidone-induced toxicity in human blood lymphocytes.

Protective Effect of Crocin against Mitochondrial Damage and Memory Deficit Induced by Beta-amyloid in the Hippocampus of Rats

Crocin is a promising preventive candidate for the potential treatment of Alzheimer’s disease and it seems that the antioxidant and neuroprotective effects of crocin are better seen when the compound is pretreated beforehand rather than introduced afterward in Aβ1-42 exposed mitochondria.

Protective effects of naringin against drugs and chemical toxins induced hepatotoxicity: A review.

In this review, available data from recent studies about the hepatoprotective effects of naringin against chemical toxicants are summarized and the possible mechanisms of actions are discussed.

Methotrexate induced mitochondrial injury and cytochrome c release in rat liver hepatocytes

It is suggested that MTX-induced cytotoxicity caused by ROS formation and GSH oxidation leads to oxidative stress and mitochondrial injury in rat hepatocytes.

Lutein Protects against Methotrexate-Induced and Reactive Oxygen Species-Mediated Apoptotic Cell Injury of IEC-6 Cells

The MTX-induced apoptosis of IEC-6 cells was shown to be repressed by the pre-treatment of lutein, which may represent a promising adjunct to conventional chemotherapy for preventing intestinal damages.

Toxicity of methyl tertiary-butyl ether on human blood lymphocytes

It was concluded that MTBE was capable of inducing oxidative stress and damage to mitochondria and lysosomes in human lymphocytes at concentrations ranging from 5 to 40 μg/L, which may be present in human blood as a result of environmental exposure.

Probucol prevents the development of CP-induced cardiotoxicity by a mechanism related, at least in part, to its ability to increase mRNA expression of antioxidant genes and to decrease apoptosis in cardiac tissues with the consequent improvement in mitochondrial oxidative phosphorylation and energy production.

Evaluation of cyclophosphamide-induced genotoxicity and cytotoxicity in cultured human lymphocytes

CP has genotoxic and cytotoxic effect on cultured human lymphocytes as compared to normal, and the frequency of micronuclei in binucleated cells showed a dose-dependent increase.

Age associated oxidative damage in lymphocytes

The results indicated that, balance of oxidant and antioxidant systems in lymphocytes shifts in favor of accelerated oxidative damage during aging, Thus oxidative stress in lymphocyte may particular interest in aging and may play important role in immunosenescence.

Mitochondrial Permeability Transition and Cell Death: The Role of Cyclophilin D

Direct and indirect mechanisms underlying CyP-D interaction with a target protein of the mPTP complex are discussed, which will help to further develop new pharmacological agents targeting mitochondria-mediated cell death.
