Mitochondria are dynamic intracellular organelles that play a central role in oxidative metabolism and apoptosis. The recent resurgence of interest in the study of mitochondria has been fuelled in large part by the recognition that genetic and/or metabolic alterations in this organelle are causative or contributing factors in a variety of human diseases including cancer. Several distinct differences between the mitochondria of normal cells and cancer cells have already been observed at the…

Cancer — the mitochondrial connection
TLDR
Biochemical and biophysical characterization have helped to identify several important differences between mitochondria of normal and disease state and such unique alterations in mitochondrial structure and function could become promising target for the development of new generation drugs.
Mitochondrial DNA: The Alternative Molecular Marker for Cancer Detection and Treatment
TLDR
This review suggests that mitochondrion is one such target that might prove clinically useful either as markers for the early detection of cancer or as unique molecular sites against which novel and selective chemotherapeutic agents might be targeted.
Small mitochondria-targeting molecules as anti-cancer agents.
Shutting Down the Furnace: Preferential Killing of Cancer Cells with Mitochondrial-Targeting Molecules.
TLDR
Several targeting strategies aimed at delivering useful cargo to mitochondria, including the use of delocalized lipophilic cations coupled to useful molecules, or peptides that insert in mitochondrial membranes are reviewed.
Mitochondria as targets for cancer therapy.
TLDR
Mitochondria have recently emerged as intriguing targets for anticancer drugs, and a variety of compounds have been now identified that act via mitochondria, which can be expected to be very efficient drugs of choice for a number of different types of the neoplastic disease.
Recent Progress in Mitochondria-Targeted Drug and Drug-Free Agents for Cancer Therapy
TLDR
The recent progress in mitochondria-targeted cancer therapy from the conventional approach of drug/cytotoxic agent conjugates to advanced drug-free approaches is discussed.

Mitochondrial DNA in Tumors
TLDR
Mitochondria, cellular powerhouses, play a central role in apoptosis and are suspected to contribute to aging and are suitable targets for anticancer drugs.
Mitochondria and Cancer
TLDR
The influence of mitochondria on the metastatic potential of cancer cell lines points to a promising future and in vivo characterization of populations of mitochondrialria will function as a “looking-glass” into monitoring the modulation events and predicting changes in metastatic capabilities.
Mitochondrial DNA and Inherited Diseases - A Comprehensive Review
TLDR
An overview of mitochondrial DNA structure and function is given and more specifically the metabolic and molecular alterations in mitochondria, associated with human diseases and their clinical implications are outlined.

Mitochondrial DNA determines the cellular response to cancer therapeutic agents
TLDR
Direct evidence is provided that mitDNA plays an important role in cellular sensitivity to cancer therapeutic agents by the development of resistance to adriamycin.
Somatic mutations of the mitochondrial genome in human colorectal tumours
TLDR
It is shown that mitochondria can rapidly become homogeneous in colorectal cancer cells using cell fusions and provide the first examples of homoplasmic mutations in the mtDNA of tumour cells, which have potential implications for the abnormal metabolic and apoptotic processes in cancer.
Mitochondrion as a novel target of anticancer chemotherapy.
TLDR
The design of mitochondrion-targeted cytotoxic drugs may constitute a novel strategy for overcoming apoptosis resistance.
Cancer and anticancer therapy‐induced modifications on metabolism mediated by carnitine system
TLDR
A more complex approach to mechanisms that underlie tumor growth, which takes into account the altered metabolic pathways in cancer disease, could represent a challenge for the future of cancer research.
Mitochondrial Regulation of Apoptosis
TLDR
Physiological cell death (PCD) constitutes a strictly regulated process which is responsible for the removal of superfluous, aged, or damaged cells and can result both from physiological and from pathological triggers.
Mitochondrial Biogenesis in the Liver during Development and Oncogenesis
TLDR
It is hypothesize that the aberrant mitochondrial phenotype of fast-growing hepatomas represents a reversion to a fetal program of expression of oxidative phosphorylation genes by the activation, or increased expression, of an inhibitor of β-mRNA translation.

This article reviews the major recognised toxicities associated with NRTI therapy and evidence for mitochondrial dysfunction in these complications and suggests a threshold effect for the expression of mitochondrial dysfunction, both at the mitochondrial and cellular level.

TLDR
Results demonstrate selective mitochondrial damage by MKT-077 at the cellular, biochemical, and molecular levels and suggest that selective effects on mitochondrial structure and function may provide a basis for the selective malignant cell killing exhibited by this compound.
