The chemotherapeutic value of methotrexate resides in its ability to perturb folate-dependent one-carbon metabolism and subsequently inhibit DNA synthesis. To assess the functional effect of methotrexate on hepatic one-carbon metabolism, we have developed and applied tracer kinetic techniques in vivo to quantify the carbon flux through the folate-dependent one-carbon pool in rats. Following a 7-day treatment period with methotrexate (0.2 mg/kg body weight), the tracers L-[ring-2-14C] histidine… Expand
Rat liver folate metabolism can provide an independent functioning of associated metabolic pathways
TLDR
Variation in the rates of metabolic processes associated with folate metabolism in the model revealed that folATE metabolism is organized in a striking manner that enables activation or inhibition of each individual process independently of the metabolic fluxes in others. Expand
Hepatic glycine N-methyltransferase is up-regulated by excess dietary methionine in rats.
TLDR
Data suggest that additional mechanisms contribute to regulation of GNMT such that synthesis of the protein is greater than its degradation, suggesting that the role it plays in the kidney and pancreas may in part differ from its hepatic function. Expand
Detoxification of formate by formate dehydrogenase-loaded erythrocytes and carbicarb in folate-deficient methanol-intoxicated rats.
TLDR
Formate dehydrogenase-loaded carrier erythrocytes along with carbicarb facilitates removal of formate, in methanol poisoning, in selected groups. Expand
Modulation of Methyl Group Metabolism by Streptozotocin-induced Diabetes and All-trans-retinoic Acid*
TLDR
Results indicate that both a diabetic condition and RA treatment have marked effects on the metabolism of methyl groups and homocysteine, a finding that may have significant implications for diabetics and their potential sensitivity to retinoids. Expand
Modulation of methyl group metabolism by diabetes and retinoids: implications for complications associated with a diabetic state
TLDR
The results indicate that both a diabetic condition and RA treatment cause significant alterations in the metabolism of methyl groups and homocysteine, a finding that may have implications for diabetics and the potential for complications associated with retinoid use. Expand
Mathematical modeling of folate metabolism
TLDR
The folate model is a useful platform for investigating the effects of antifolates on the folate pathway and the utility of the model is demonstrated through interrogation of drug resistance, drug‐drug interactions, drug selectivity, and drug doses and schedules. Expand
Therapeutic response to single intravenous bolus administration of formate dehydrogenase in methanol-intoxicated rats.
TLDR
It may be concluded that single bolus infusion of formate dehydrogenase facilitates fast removal of formates, a highly toxic metabolite in methanol poisoning. Expand
Effects of Taurine Deficiency and Chronic Methanol Administration on Rat Retina, Optic Nerve and Brain Amino Acids and Monoamines
TLDR
In the retina, β-Ala in combination with MTX-MeOH increased serotonin and decreased dopamine turnover rate, and resulted in changes in the amino acid balance, that could affect glycinergic activity. Expand
Effect of chronic methanol administration on amino acids and monoamines in retina, optic nerve, and brain of the rat.
TLDR
It is concluded that chronic methanol administration produced accumulation of aspartate, an excitotoxic amino acid, in the optic nerve, and this findings contribute to the understanding of meetinghanol neurotoxicity and might indicate a relationship between chronic meethanol consumption and the development of optic neuropathies. Expand
Folate regulation of axonal regeneration in the rodent central nervous system through DNA methylation.
TLDR
Folic acid and possibly other nontoxic dietary methyl donors may therefore be useful in clinical interventions to promote brain and spinal cord healing and provide possible avenues for new pharmacologic approaches to treating CNS injuries. Expand

Quantification of the carbon flow through the folate-dependent one-carbon pool using radiolabeled histidine: effect of altered thyroid and folate status.
TLDR
In the case of hyperthyroidism and dietary folate restriction, changes in static measurements such as metabolite concentrations and enzyme activities do not reflect the dynamic tracer kinetic assessment of the carbon flux that is actually occurring in vivo. Expand
Carbon flow through the hepatic folate-dependent one-carbon pool is not altered in vitamin A-deficient rats.
TLDR
It is demonstrated that vitamin A deficiency does not affect the reductive carbon flow from serine to methionine because the ability to generate methionines via remethylation of homocysteine with the carbon group originating from serines was not altered in vitamin A-deficient rats. Expand

Relationship of reduced folate changes to inhibition of DNA synthesis induced by methotrexate in L1210 cells in vivo.
TLDR
Reduced folate levels and DNA synthesis were examined in L1210 cells in mice after exposure to a wide range of methotrexate doses and sustained inhibition of DNA synthesis was generally associated with maintenance of an expanded FH2 pool and delay in repletion of the combined tetrahydrofolate pools. Effect of nitrous oxide inactivation of vitamin B12-dependent methionine synthetase on the subcellular distribution of folate coenzymes in rat liver.
TLDR
The results show that the effects of nitrous oxide inactivation of vitamin B12 are confined to the cytosol, at least in the short term, and suggest that there is little, if any, transport of free folates between the cytOSolic and mitochondrial compartments. Expand
Effects of choline deficiency and methotrexate treatment upon rat liver.
TLDR
The hypothesis that the fatty infiltration of the liver associated with MTX treatment occurs because of a disturbance in choline metabolism is consistent with the hypothesis that choline deficiency and treatment with methotrexate are related. Expand
The effect of methotrexate on intracellular folate pools in human MCF-7 breast cancer cells. Evidence for direct inhibition of purine synthesis.
TLDR
The rapid inhibition of de novo purine synthesis in these cells following methotrexate exposure coupled with a relatively preserved 10-formyl-H4PteGlu pool suggests direct inhibition of this synthetic pathway by the temporally coincident accumulation of dihydrofolic acid and/or metotrexate polyglutamates.
