Effect of methotrexate (MTX) on mitochondrial oxygen uptake, oxidative phosphorylation and on the activity of several enzymes linked to respiratory chain was studied. MTX was able to inhibit state III respiration activated by ADP and to decrease the respiratory coefficient with the substrates α‐ketoglutarate and glutamate; these effects became pronounced when mitochondria were pre‐incubated with MTX for 10 min. No effect was observed on ATPase activity of undamaged or broken mitochondria; the… Expand
Methotrexate: Studies on cellular metabolism. II—Effects on mitochondrial oxidative metabolism and ion transport
TLDR
MTX has an inhibitory effect on the activities of the oxidation enzymes of NAD+‐linked substrates without interfering with the oxidation systems of FAD‐ linked substrates. Expand
Methotrexate: pentose cycle and oxidative stress
TLDR
The net results appear to show that the biological situation resulting from treatment with MTX leads to a reduction of effectiveness of the antioxidant enzyme defence system. Expand
Effect of methotrexate (MTX) on NAD(P)+ dehydrogenases of HeLa cells: malic enzyme, 2‐oxoglutarate and isocitrate dehydrogenases
TLDR
An evaluation of the effects of MTX on malic enzyme activity as measured by pyruvate plus lactate production in intact cells supplied with malate showed a decrease of about 40 per cent in metabolite production using 0·4 mM MTX. Expand
Methotrexate use and NAD+/NADH metabolism in psoriatic keratinocytes
TLDR
As expected, the topical application of NAD+ (oxidized form of nicotinamide-adenine dinucleotide) resulted in a clinical improvement of psoriasis lesions in one study, Nevertheless, another report revealed reduced fluorescence of NADH in psoriatic plaques. Expand
Methotrexate: Studies on cellular metabolism. III.‐Effect on the transplasma‐membrane redox activity and on ferricyanide‐induced proton extrusion by HeLa cells
TLDR
It is suggested that inhibition of electron transport and proton extrusion caused by MTX could be associated with other metabolic alterations such as response to the increase in NADH levels and decrease in intracellular pH. Expand
Central role of mitochondria in drug-induced liver injury
TLDR
Susceptibility to parent drug-mediated mitochondrial dysfunction can be increased by factors impairing the removal of the toxic parent compound or by the presence of other medical condition(s) impairing mitochondrial function. Expand
A study on the possible role of mitochondrial damage reactive nitrogen species and reactive oxygen species in the pathogenesis of methotrexate induced small intestinal damage in the rat
Methotrexate is widely used in the treatment of different malignancies and inflammatory disorders. The therapeutic effect of the drug is through the competitive inhibition of dihydrofolateExpand
Drug-induced toxicity on mitochondria and lipid metabolism: mechanistic diversity and deleterious consequences for the liver.
TLDR
In obese and diabetic patients, some drugs may induce acute liver injury more frequently while others may worsen the pre-existent steatosis (or steatohepatitis), which is characterized not only by lipid accumulation but also by necroinflammation and fibrosis. Expand
Deficiency of the Mitochondrial NAD Kinase Causes Stress-Induced Hepatic Steatosis in Mice.
TLDR
In mice given injections of methotrexate, supplementation of a diet with nicotinamide riboside, an NAD precursor, replenished hepatic NADP and protected the mice from hepatotoxicity, based on markers such as increased level of serum alanine aminotransferase. Expand

TLDR
The current review highlights the crucial role that FPGS plays in maintenance of folate homeostasis under physiological conditions and delineates the plethora of the molecular mechanisms underlying loss of FPGs function and consequent antifolate resistance in cancer. Expand
