Rat liver cells, isolated by chelate perfusion and extrusion through a tissue press, incorporated labeled acetate into cellular lipids and into lipids released into the suspending medium. Optimal rates of incorporation required supplementation of tris-KCl medium with Mg++, Mn++, succinate, citrate, nicotinamide, Coenzyme A, NADP and glucose-6-phosphate.
