Alternative pre-mRNA splicing is a central mode of genetic regulation in higher eukaryotes. Variability in splicing patterns is a major source of protein diversity from the genome. In this review, I describe what is currently known of the molecular mechanisms that control changes in splice site choice. I start with the best-characterized systems from the Drosophila sex determination pathway, and then describe the regulators of other systems about whose mechanisms there is some data. How these…
Alternative pre-mRNA splicing in the human system: unexpected role of repetitive sequences as regulatory elements
TLDR
The role of short sequence repeats, which are often polymorphic, in splicing regulation is discussed, and the underlying mechanistic principles, focussing on the cis-acting regulatory elements, are summarized.
Systems analysis of alternative splicing and its regulation
TLDR
Recent progress in systems biology approaches are starting to define how combinatorial splicing regulation shapes the complex splicing phenotypes observed in different tissue types and developmental stages and under different conditions.
Mechanisms of alternative splicing regulation: insights from molecular and genomics approaches
TLDR
Great progress has been made by studying individual transcripts and through genome-wide approaches, which together provide a better picture of the mechanistic regulation of alternative pre-mRNA splicing.
