Thromboxane synthase (TS) catalyzes the conversion of the prostaglandin endoperoxide into thromboxane A2 (TxA2), a potent vasoconstrictor and inducer of platelet aggregation. In concert with prostacyclin TxA2 plays a pivotal role in the maintenance of hemostasis. Deficiency of platelet TS activity has been shown to result in bleeding disorders. The potent effect of TxA2 on platelet function and vascular activity suggests a possible involvement of TS in normal and pathophysiological conditions… Expand
Genomic organization, chromosomal localization, and expression of the murine thromboxane synthase gene.
TLDR
These studies provide the genetic tools and information for TS research in mice, which should expedite understanding of the genetic contribution of TS in normal physiology as well as in disease states. Expand
Genomic structure and polymorphism of the human thromboxane synthase-encoding gene.
TLDR
Southern analysis indicates that the human haploid genome contains a single copy of the TS gene, and transcription of hTS is primarily TATA-independent, as determined by promoter-directed reporter gene expression in transfected cells. Expand
Functional analysis of human thromboxane synthase polymorphic variants
TLDR
The increased Km and decreased Vmax values observed with L357V suggest that this variant may generate less TXA2 at the low levels of PGH2 expected in vivo, raising the possibility of attenuated signaling through the thromboxane pathway. Expand
Characterization of the human gene (TBXAS1) encoding thromboxane synthase.
TLDR
A tissue-distribution study demonstrated that thromboxane synthase mRNA is widely expressed in human tissues and is particularly abundant in peripheral blood leukocyte, spleen, lung and liver. Expand
Association analysis of thromboxane A synthase 1 gene polymorphisms with aspirin intolerance in asthmatic patients.
TLDR
The rare allele of rs6962291 in intron 9 was significantly lower in the AIA group than in the ATA group and may play a protective role against aspirin hypersensitivity via a lower catalytic activity of the TBXAS1 gene. Expand
Association of thromboxane A1 synthase (TBXAS1) gene polymorphism with acute urticaria induced by nonsteroidal anti-inflammatory drugs.

Genetic analysis of factor XII and bradykinin catabolic enzymes in a family with estrogen-dependent inherited angioedema and the effect of androgen prophylaxis on plasma aminopeptidase P determines optimal conditions for activating prekallikrein. Expand
The porcine thromboxane synthase-encoding cDNA: sequence, mRNA expression and enzyme production in Sf9 insect cells.
TLDR
A high-titer recombinant baculovirus containing pTS cDNA was developed to conduct a time course study of enzyme production in Spodoptera frugiperda (Sf9) cells and produced TS resembles purified pTS in catalysis, as well as inhibition by a substrate analog inhibitor. Expand
Effects of the flavonoid biochanin A on gene expression in primary human hepatocytes and human intestinal cells.
TLDR
BCA treatment up-regulated genes mainly in the CYP2 family, which can influence the metabolism of xenobiotics, producing effects of pharmacological and toxicological importance. Expand
Effects of copy number variable regions on local gene expression in white blood cells of Mexican Americans
TLDR
It is found that genes whose expression level is significantly influenced by nearby CNVRs are enriched for immunity and autoimmunity related genes, and CNVs that directly overlap gene sequences have the largest effects on gene expression. Expand
Assigning single clinical features to their disease-locus in large deletions: the example of chromosome 1q23-25 deletion syndrome
TLDR
To overcome the problem of incomplete penetrance, an algorithm is developed that is applied to the deletion region 1q23.3-q25, which contains three SROs, each contributing to the abnormal phenotype without clearly distinguishing between the different malformations. Expand
