Lysyl hydroxylases : characterization of mouse lysyl hydroxylases and generation of genetically modified lysyl hydroxylase 3 mouse lines

  title={Lysyl hydroxylases : characterization of mouse lysyl hydroxylases and generation of genetically modified lysyl hydroxylase 3 mouse lines},
  author={Heli Ruotsalainen},
Lysyl hydroxylase (EC, procollagen-lysine, 2-oxyglutarate, 5-dioxygenase, Plod) catalyzes the hydroxylation of certain lysine residues in collagens and in other proteins with collagenous domains. Three lysyl hydroxylase isoforms have been cloned from human and rat. The importance of lysyl hydroxylase 1 in collagen biosynthesis is demonstrated by the heritable disorder, Ehlers-Danlos syndrome type VI, which is characterized by joint laxity, progressive scoliosis, muscle hypotonia…

Expanding the lysyl hydroxylase toolbox: New insights into the localization and activities of lysyl hydroxylase 3 (LH3)
It is shown that growth cone migration depends critically on the LH3 glycosyltransferase domain, but, unlike the other isoforms, LH3 is also found in the extracellular space in some tissues, suggesting that it may have a role in matrix remodeling.

Novel lysyl hydroxylase isoforms
Novel human lysyl hydroxylases, 2a, 2b and 3 isoforms, were characterized and all these novel isoforms were able to hydroxyate lysine residues in short collagenous peptides.

Lysyl Hydroxylase 3 Is a Multifunctional Protein Possessing Collagen Glucosyltransferase Activity*
In vitro mutagenesis experiments demonstrate that the amino acids involved in the GGT active site differ from those required for LH3 activity, and antibodies recognizing the amino acid sequence of human LH3 and those against a highly purified chicken GGT partially inhibited the G GT activity.
Failure of highly purified lysyl hydroxylase to hydroxylate lysyl residues in the non-helical regions of collagen.
Analysis of chick type I collagen products obtained following bacterial collagenase digestion of the reaction mixture showed that overall 37% hydroxylation of lysyl residues within the helical region of collagen had been obtained, which may be maximal, providing strong circumstantial evidence for the suggestion that a lysym hydroxymase specific for the telopeptidyl residues and distinct from that active towards lysy residues in the helicals portion of the molecule may exist.

Lack of collagen type specificity for lysyl hydroxylase isoforms.
It is suggested that LH1, LH2, and the alpha subunit of prolyl 4-hydroxylase are coregulated together with total collagen synthesis but not with the specific collagen types and indicate that LH3 behaves differently from LH1 and LH2 , implying a difference in their substrates.
