Ling Zhi-8 (LZ-8), an immunomodulatory protein, is derived from and has been cloned from the medicinal mushroom Ganoderma lucidum (Reishi or Ling Zhi); this protein exhibits immunomodulating and antitumor properties. We investigated the effects of recombinant LZ-8 protein (rLZ-8) on the proliferation of A549 human lung cancer cells. Here, we showed that rLZ-8 inhibits cell growth and that this is correlated with increased G(1) arrest. The treatment of A549 cells with rLZ-8 activated p53 and p21…

Ling-Zhi-8 protein (LZ-8) suppresses the production of pro-inflammatory mediators in murine microglial BV-2 cells

LZ-8 reduced TLR-4-mediated NF-κB translocation into nucleus resulting in the suppression of pro-inflammatory mediator expression and production in microglial cells.

The oncoprotein HBXIP promotes human breast cancer growth through down-regulating p53 via miR-18b/MDM2 and pAKT/MDM2 pathways

The molecular mechanisms underlying the modulation of MDM2/p53 interaction by HBXIP in human breast cancer MCF-7 cells in vitro and in vivo are investigated to shed light on a novel mechanism of p53 down-regulation during the development of breast cancer.

Small Ribosomal Protein Subunit S7 Suppresses Ovarian Tumorigenesis through Regulation of the PI3K/AKT and MAPK Pathways

The results suggest that RPS7 suppresses ovarian tumorigenesis and metastasis through PI3K/AKT and MAPK signal pathways, and may be used as a potential marker for diagnosis and treatment of ovarian cancer.

GMI, an Immunomodulatory Protein from Ganoderma microsporum, Potentiates Cisplatin-Induced Apoptosis via Autophagy in Lung Cancer Cells.

This is the first study to reveal the novel function of GMI in potentiating cisplatin-mediated apoptosis and suggest that GMI may be a potential cis platin adjuvant against lung cancer.

Recent progress in understanding the molecular basis of p53 signaling, particularly the newly identified ribosomal stress—p53 pathway, is reviewed and the development of chemotherapeutics via activating wild-type p53 or restoring mutant p53 functions in cancer is reviewed.

Induction of Cbl‐dependent epidermal growth factor receptor degradation in Ling Zhi‐8 suppressed lung cancer

The first evidence showing that rLZ‐8 inhibits growth and induces apoptosis of lung cancer cells by promoting EGFR degradation is provided, suggesting a novel anti‐cancer function of rLz‐8 that targeting EGFR overexpression or mutation as well as EGFR‐dependent processes in cancer cells.

Correlation of the Structure and Bioactivity of Recombinant Fungal Immunomodulatory Protein, Ling-Zhi-8 (LZ-8) Following Exposure to Denaturing Conditions

Investigation of the effects of exposure of LZ-8 to denaturing conditions on its ability to suppress inflammatory responses in lipopolysaccharide-stimulated RAW 264.7 macrophages found this effect was not dependent on the native protein structure, suggesting that the entire protein might not be responsible for the bioactive properties of Lz-8.

Involvement of Bcl-X(L) deamidation in E1A-mediated cisplatin sensitization of ovarian cancer cells.

It is found that the E1A transfectants became sensitive to cisplatin-induced apoptosis compared to the parental cells, SKOV3.ip1 cells, and deamidation of the deamidated forms of Bcl-X(L) contributes to E 1A-mediated cisPlatin sensitization in SKov3.IP1 cells.

Ling Zhi-8: a novel T cell mitogen induces cytokine production and upregulation of ICAM-1 expression.

It is demonstrated that LZ-8 is a potent T cell activator, mediating its effects via cytokine regulation of integrin expression, as well as a protein purified from Ganoderma lucidium thought to possess potent effects on the immune system.

Inhibitory effects of ganoderma lucidum on tumorigenesis and metastasis of human hepatoma cells in cells and animal models.

The results suggest that the lucidenic acid-rich G. lucidum extract could serve as a chemopreventive agent for the tumorigenesis and metastasis of highly invasive hepatoma cells.

Ganoderma lucidum inhibits proliferation and induces apoptosis in human prostate cancer cells PC-3.

The data demonstrate that Ganoderma lucidum inhibits cell proliferation in a dose- and time-dependent manner by the down- regulation of expression of cyclin B and Cdc2 and by the up-regulation of p21 expression, and exerts its effect on cancer cells by multiple mechanisms.

Involvement of Bcl-XL deamidation in E1A-mediated cisplatin sensitization of ovarian cancer cells

It is found that the E1A transfectants became sensitive to cisplatin-induced apoptosis compared to the parental cells, SKOV3.ip1 cells, and this findings suggest that Bcl-XL deamidation contributes to E 1A-mediated cisPlatin sensitization in SKOV1 cells.

LING ZHI‐8: STUDIES OF A NEW IMMUNOMODULATING AGENT

LZ-8 proved to have a significant effect on cellular immunity, since its administration in an allografted mouse skin model resulted in an increased survival time, and in a model of transplanted allogeneic pancreatic rat islets, Lz-8 was effective in delaying the rejection process of allogsrafted islets.

Ribosomal Protein L11 Negatively Regulates Oncoprotein MDM2 and Mediates a p53-Dependent Ribosomal-Stress Checkpoint Pathway

It is shown that the functional consequence of L11-HDM2 association results in the prevention of HDM2-mediated p53 ubiquitination and degradation, subsequently restoring p53-mediated transactivation, accumulating p21 protein levels, and inducing a p 53-dependent cell cycle arrest by canceling the inhibitory function of HDm2.

Ganoderma lucidum extract induces cell cycle arrest and apoptosis in MCF‐7 human breast cancer cell

The alcohol extract of Ganoderma lucidum inhibited cell proliferation in a dose‐ and time‐dependent manner and can directly induce apoptosis in MCF‐7 cells, which might be mediated through up‐regulation of p21/Waf1 and down‐ regulation of cyclin D1.
