}The relationship between acute toxicity in rats (LD50 values) and indicators of potential health hazards in humans was investigated, based on a chemical population-based paradigm (i.e. the “chemical diversity approach”). These structure–activity relationship-based analyses indicate that high toxicity in rats (i.e. a low LD50 value) is not a good predictor of health effects in humans. In fact, it was found that high acute toxicity to minnows, as well as toxicity to cultured cells, showed…

Comparative Quantitative Structure–Activity–Activity Relationships for Toxicity to Tetrahymena pyriformis and Pimephales promelas
- ChemistryAlternatives to laboratory animals : ATLA
- 2007
An approach for predicting acute aquatic toxicity, in the form of a quantitative structure–activity–activity relationship (QSAAR), is described, and structural features that were found to improve the extrapolation between the toxicity to the two different species were related to the electron distribution of the carbon skeleton of the toxicant.
Extrapolating from experimental to human studies via the in vitro paradigm

A previously unknown metabolic capability of urothelial cells that can be modelled ex vivo is identified, and fundamental differences in rat and human u rothelial cell physiology are demonstrated that support the validation of human tissue-specific in vitro models for assessment of xenobiotic toxicity.

It is demonstrated that neural network methods can be utilized to rank compounds according to their relative toxicity using COPAS-generated data obtained from exposing a large number of nematodes to water-soluble compounds.
Priority setting strategies for regulatory testing of industrial chemicals

For the majority of the estimated 70,000 industrial chemical substances available on the European market today there is not enough information to enable a reasonably complete assessment of the risk to public health, according to the European Chemicals Agency.

The project described here aims to address the initial phase ofbasic cytotoxic effects of cosmetic ingredients following dermal exposure, by using human skin cells (dermal fibroblasts) as a relevant model for the initial local response, and careful extrapolation to the basic cytotoxicity likely to occur at other eventual exposure sites in the human body.

An advanced in vitro method, which can significantly reduce the number of animals needed for the toxicity testing of a broad range of compounds/xenobiotics, is presented, ready for validation, and could then be incorporated into OECD guidelines, thus reducing the total number ofAnimals needed for regulatory toxicity testing.
A Possible Relationship Between Toxicity and Carcinogenicity
- Medicine
- 1986
Comparisons of acute toxicity (LD50) with carcinogenic potency show that they are correlated, but the correlation may in part be an artifact, since doses used in the NCI/NTP carcinogenesis bioassays are toxic and because reliable measures of potency can only be derived for positive carcinogenic responses.

The analyses revealed that there was a structural basis for the MTD in mice and in rats and that these overlapped considerably and there was also a significant “antagonism” between such fragments; i.e., fragments associated with high toxicity (low MTD) were associated with lack of carcinogenicity and vice versa.
