Biglycan is an important component of the extracellular matrix, and it is also a member of small leucine-rich proteoglycan family. Previous studies indicated that the expression of biglycan was increased in a variety of tumor tissues, including colon cancer. However, the mechanisms underlying its effects in colon cancer remain to be fully elucidated. In the present study, the effects of biglycan knockdown on colon cancer cell proliferation, migration, invasion and apoptosis were investigated…

Biglycan promotes the chemotherapy resistance of colon cancer by activating NF-κB signal transduction
TLDR
Findings showed that BGN contributed to the formation of chemotherapy resistance in colon cancer cells by activating NF-κB signaling.
Lumican mediates HTB 94 chondrosarcoma cell growth via an IGF ‐ IR / Erk 1 / 2 axis
TLDR
The findings of the present study demonstrate that endogenous lumican is a novel regulator of HTB94 cell growth through an IGF-IR/ERK1/2/p53 signaling cascade.
Biglycan Promotes Cancer Stem Cell Properties, NFκB Signaling and Metastatic Potential in Breast Cancer Cells
Simple Summary Breast cancer stem cells (BCSCs) are a small sub-population of cells within tumors with high metastatic potential. We identified biglycan (BGN) as a prospective molecular target in
Biglycan, a Nitric Oxide-Downregulated Proteoglycan, Prevents Nitric Oxide-Induced Neuronal Cell Apoptosis via Targeting Erk1/2 and p38 Signaling Pathways
TLDR
It is reported that BGN protects against NO-induced apoptosis in human neuroblastoma SH-EP1 cells, and downregulation of BGN by siRNA aggravated No-induced neuronal cell death, which was not attenuated by U0126 or SB203580.
The Role of Decorin and Biglycan Signaling in Tumorigenesis
TLDR
Growing evidence for the complex roles of decorin and biglycan signaling in tumor biology is summarized and potential novel therapeutic implications are addressed.
Altered expression of small leucine-rich proteoglycans (Decorin, Biglycan and Lumican): Plausible diagnostic marker in urothelial carcinoma of bladder
TLDR
Higher expression of Lumican in Bladder cancer patients with the cut-off value of highest optimum sensitivity and specificity shows its importance as a potential non-invasive marker for early detection of UBC following further validation in large patient cohort.
The Genes Encoding Small Leucine-Rich Proteoglycans Undergo Differential Expression Alterations in Colorectal Cancer, Depending on Tumor Location
TLDR
In vitro studies using the HT29 tumor cell line suggest that deregulation of SLRPs could affect cell proliferation, and small leucine-rich proteoglycans constitute new differential markers of RS- and LSCCs, showing differences dependent on the anatomical location of the tumor.

Biglycan up-regulated vascular endothelial growth factor (VEGF) expression and promoted angiogenesis in colon cancer
TLDR
Biglycan-mediated VEGF regulation may correlate with the activation of the ERK signaling pathway, and biglycan may be a promising target for anti-angiogenic therapy for cancer.
Biglycan is overexpressed in pancreatic cancer and induces G1-arrest in pancreatic cancer cell lines.
TLDR
The ECM protein PG-I inhibits growth by arresting pancreatic cancer cells in G1 and may be part of a host defense mechanism aimed at slowing down pancreatic tumor progression.
HER-2/neu-mediated Down-regulation of Biglycan Associated with Altered Growth Properties*
TLDR
Using an in vitro model of oncogenic transformation, a down-regulation of Bgn expression as well as an altered secretion of different Bgn isoforms was found both in murine and human HER-2/neu oncogene-transformed cells when compared with HER- two-neu− cells, associated with a reduced growth, wound closure, and migration capacity.
Biglycan expression correlates with aggressiveness and poor prognosis of gastric cancer
TLDR
Elevated expression of biglycan may play an important role in the development and progression of gastric cancer, and could be further evaluated as a biomarker for predication of a poor clinical outcome.
Up-regulated biglycan expression correlates with the malignancy in human colorectal cancers
TLDR
Biglycan mRNA expression was analyzed in 110 samples derived from 55 patients with colorectal cancer using quantitative real-time RT-PCR and showed a significant correlation in biglycan up-regulation with poor tumor differentiation, lymph node metastasis, and distant metastasis.
Biglycan expression and clinical outcome in patients with pancreatic adenocarcinoma
TLDR
Assessing the prognostic role of biglycan expression in pancreatic cancer confirmed PG-I expression and MIB-1 as independent negative prognostic factors and underline the complexity of tumor–stroma interactions in the advanced stage of cancer and the need of further study.
Tumor cell interactions with the extracellular matrix during invasion and metastasis.
TLDR
Preliminary findings suggest that cell-matrix interactions influence gene expression and that the protease inhibitor balance can greatly influence cell-Matrix interactions, so it appears that all three steps in the invasive process are linked and interdependent.
Quantitative proteomics analysis integrated with microarray data reveals that extracellular matrix proteins, catenins, and p53 binding protein 1 are important for chemotherapy response in ovarian cancers.
TLDR
The results suggest that chemotherapy response may be determined by multiple and complex system properties involving extracellular-matrix, cell adhesion and junction proteins.
Opposing Effects of ERK and JNK-p38 MAP Kinases on Apoptosis
TLDR
The effects of dominant-interfering or constitutively activated forms of various components of the JNK-p38 and ERK signaling pathways demonstrated that activation of JNK and p38 and concurrent inhibition of ERK are critical for induction of apoptosis in these cells.
Large matrix proteoglycans, versican and perlecan, are expressed and secreted by human leukemic monocytes.
TLDR
The production of these large matrix PGs by THP-1 monocytes is reported for the first time and may be of importance in monocyte malignant transformation and differentiation.
