Allosteric enzymes have very complex kinetic behaviours which are primarily interpreted through simplified models. To describe the functional properties of liver and muscle glycogen phosphorylase isozymes we have developed an experimental strategy based on the measurements of initial reaction rates in the presence of different concentrations of the effectors glucose‐1‐phosphate and methyl‐xanthines. Using the extensive structural information available for the two glycogen phosphorylase…
Investigations on the mechanism of allosteric activation of rabbit muscle glycogen phosphorylase b by AMP
TLDR
This study has employed linkage analysis in combination with structural perturbations via site-directed mutagenesis to test kinetic models of activation of phosphorylase b by AMP, and to examine the roles of the N-terminus, the acidic patch, α-helix 1 and the 280’s loop in activation byAMP.

TLDR
It was shown that a GPa-rich extract of rat liver contained GSH-dependent As(V)-reducing activity that was inhibited by specific GPIs, suggesting that the liver-type GPa can also catalyze reduction of As( V), supporting that the latter is coupled to glycogenolysis.
Hepatic 11 beta-hydroxysteroid dehydrogenase 1 involvement in alterations of glucose metabolism produced by acidotic stress in rat
TLDR
The results demonstrate the involvement of HSD1 in stress induced carbohydrate disturbances and could contribute to the impact of HDS1 inhibitors on carbohydrate metabolism and its relevance in the study of Metabolic Syndrome Disorder and non insulin-dependent diabetes mellitus.

Synergistic regulation of phosphorylase a by glucose and caffeine.
TLDR
Kinetic studies of both liver and muscle phosphorylase a demonstrate that caffeine and glucose inhibit the binding of glucose 1-phosphate to the enzyme in a synergistic competitive and nonexclusive manner, suggesting that glucose homeostasis as regulated by phosphoryLase may be dependent on a second ligand and that the role of glucose in this physiological process may have been overestimated.
Thermodynamic Characterization of 5′-AMP Binding to Bovine Liver Glycogen Phosphorylase a *
TLDR
The binding of adenosine 5′-monophosphate to liver glycogen phosphorylase a has been studied by size exclusion high performance liquid chromatography and isothermal titration microcalorimetry at pH 6.9 over a temperature range of 25 to 35°C and shows that protons are released during the binding of the nucleotide.

TLDR
The allosteric mechanism of activation of phosphorylase by phosphorylation may be relevant to other enzymes although it is now known that other mechanisms such as electrostatic steric blocking mechanisms also exist.
Analogous activation of bovine liver glycogen phosphorylase by AMP and IMP.
The nature of the binding site for aromatic compounds in glycogen phosphorylase b.
TLDR
The inhibition of rabbit muscle glycogen phosphorylase b by aromatic compounds was examined and the relative effectiveness of the inhibitors correlated well with increasing substituent constant, pi, indicating the hydrophobic nature of the binding site.

Structural basis of the synergistic inhibition of glycogen phosphorylase a by caffeine and a potential antidiabetic drug.
TLDR
The complex structure provides a rationale to understand the structural basis of the synergistic inhibition between W1807 and caffeine and induces substantial conformational changes both in the vicinity of the allosteric site and the subunit interface which transform GPa to the T'-like state conformation already observed with GPa-glucose-W1807 complex.
Structural relationships among regulated and unregulated phosphorylases.
TLDR
Comparing crystallographic structures of regulated and unregulated phosphorylases, including maltodextrinosphorylase (MalP) from Escherichia coli, glycogen phosphoryLase from yeast, and mammalian isozymes from muscle and liver tissues, provides insights into the structural basis for allosteric control mechanisms.
