Isolation of nuclei is widely used in analyses such as DNA footprinting, gel retardation and runon transcription assays. When we applied the NP-40 lysis method of Nevins (1) to progenitors of the monocytic lineage, we found that these cells have a very fragile nuclear membrane which is easily susceptible to partial premature lysis.
