PURPOSE
To isolate and characterize goblet cells from normal human conjunctival tissue to determine whether epidermal growth factor (EGF) receptors are present and whether EGF can influence goblet cell proliferation.
METHODS
Goblet cells were isolated from explant cultures established from normal conjunctival tissue harvested from patients during periocular surgery. The cells were grown in RPMI culture medium supplemented with 10% fetal bovine serum and characterized using morphology…

An in vitro organ culture method to investigate human conjunctival epithelial basal precursor cells and their progeny within a more natural three-dimensional microenvironment provides new opportunities to investigate epithelial reconstitution of the Conjunctival surface and changes that may have occurred to their stem/precursor cells during adaptation to varying conditions in vitro.

The serum-free-derived epithelial equivalents demonstrated a significant increase in proliferation and stratification after transplantation, which is crucial for enhancing graft survival and regeneration of the conjunctival surface after clinical transplantation.

The simulation of a niche environment in vitro by co-culturing mitotically active subconjunctival fibroblasts with conjunctival epithelial cells supports the maintenance of conjunctiver cells with progenitor cell characteristics and therefore might be a useful tool to expand conjunctiva epithelial progenitors cells in vitro for clinical use.

The proliferative capacity of conjunctival epithelial cells cultivated in HS supplemented cultures was comparable to FBS and BPE supplemented cultures, indicating the elimination of animal material from the culture system is advantageous when cultivating cells for clinical transplantation.

It is concluded that uninjured, adult LG contains progenitor cells that may be MECs, which can be isolated and differentiated into multiple lineages.
