Physical exercise is able to improve skeletal health. However, the mechanisms are poorly known. Irisin, a novel exercise-induced myokine, secreted by skeletal muscle in response to exercise, have been shown to mediate beneficial effects of exercise in many disorders. In the current study, we demonstrated that irisin promotes osteoblast proliferation, and increases the expression of osteoblastic transcription regulators, such as Runt-related transcription factor-2, osterix/sp7; and osteoblast…

TLDR
The mechanism responsible for the therapeutic potential of irisin in bone metabolism disease is revealed and it is shown that irisin promotes proliferation but inhibits differentiation in osteoclast precursor cells.
Irisin promotes cementoblast differentiation via p38 MAPK pathway.
TLDR
Irisin can promote the differentiation of cementoblasts via p38 MAPK pathway through its role in promoting glucose metabolism and cortical bone mass and strength.
The immunomodulatory role of irisin on osteogenesis via AMPK-mediated macrophage polarization.
Irisin promotes C2C12 myoblast proliferation via ERK-dependent CCL7 upregulation
TLDR
The results suggest that irisin promotes C2C12 myoblast proliferation via ERK-dependent CCL7 upregulation and may aid in understanding how irisin contributes to muscle development.
Irisin Recovers Osteoarthritic Chondrocytes In Vitro
TLDR
It is suggested that irisin may stimulate hOAC proliferation and anabolism inhibiting catabolism through p38, Akt, JNK, and NFκB inactivation in vitro, demonstrating the existence of a cross-talk between muscle and cartilage.
The Myokine Irisin Promotes Osteogenic Differentiation of Dental Bud-Derived MSCs
TLDR
The myokine irisin, well known for its anabolic effect on bone tissue, has been demonstrated to positively act on osteoblastic differentiation processes in vitro, and could improve bone tissue metabolism in MSC regenerative procedures.
Myokine Irisin promotes osteogenesis by activating BMP/SMAD signaling via αV integrin and regulates bone mass in mice
TLDR
In vitro study showed that lack of irisin impeded the MSC-derived osteogenesis of FNDC5-/- mice, and in vitro findings further indicate that irisin regulates bone homeostasis.
Recombinant Irisin Prevents the Reduction of Osteoblast Differentiation Induced by Stimulated Microgravity through Increasing β-Catenin Expression
TLDR
This study is the first to show that r-irisin positively regulates osteoblast differentiation under simulated microgravity through increasing β-catenin expression, which may reveal a novel mechanism, and it provides a prevention strategy for bone loss and muscle atrophy induced by microgravity.
The isoflavone genistein enhances osteoblastogenesis: signaling pathways involved
TLDR
Genistein promoted osteoblastogenesis through the participation of ER and NOS pathways, and the contribution of ERK or PI3K signal transduction pathways, and also stimulates osteoclast differentiation from its mononuclear progenitor.

Irisin Enhances Osteoblast Differentiation In Vitro
TLDR
New evidence supports the idea that muscle can be considered an endocrine organ, given its ability to target adipose tissue by promoting energy expenditure, and shows, for the first time, that Irisin directly targets osteoblasts, enhancing their differentiation.
A Specific Oligodeoxynucleotide Promotes the Differentiation of Osteoblasts via ERK and p38 MAPK Pathways
TLDR
Results suggest that the ERK1/2 and p38 MAPK pathways, as well as Runx2 activation, are involved in ODN MT01-induced up-regulation of osteocalcin, type I collagen and the activity of ALP in MG 63 cells.
Irisin Promotes Human Umbilical Vein Endothelial Cell Proliferation through the ERK Signaling Pathway and Partly Suppresses High Glucose-Induced Apoptosis
TLDR
It is suggested that irisin plays a novel role in sustaining endothelial homeostasis by promoting HUVEC proliferation via the ERK signaling pathway and protects the cell from high glucose-induced apoptosis by regulating Bcl-2,Bax and Caspase expression.
Spata4 promotes osteoblast differentiation through Erk‐activated Runx2 pathway
TLDR
The spermatogenesis associated 4 gene is expressed in osteoblasts and that overexpression of Spata4 accelerates osteoblast differentiation and mineralization in MC3T3‐E1 cells, providing new evidence that Spata 4 plays a role in regulation of osteOBlast differentiation.
Chrysin promotes osteogenic differentiation via ERK/MAPK activation
TLDR
The effect of chrysin on osteogenesis is ERK1/2-dependent and involves ER and has the significant potential to enhance osteogenesis for osteoporosis prevention and treatment.
Irisin Stimulates Browning of White Adipocytes Through Mitogen-Activated Protein Kinase p38 MAP Kinase and ERK MAP Kinase Signaling
TLDR
The data suggest that irisin can potentially prevent obesity and associated type 2 diabetes by stimulating expression of WAT browning-specific genes via the p38 MAPK and ERK pathways.
Irisin stimulates muscle growth-related genes and regulates adipocyte differentiation and metabolism in humans
TLDR
In human adipocytes, irisin induced uncoupling protein 1 and consequently increased adipocyte energy expenditure, expression of metabolic enzymes and metabolite intermediates, resulting in inhibition of lipid accumulation, suggesting an additional mechanism in suppressing fat mass.
Regulatory controls for osteoblast growth and differentiation: role of Runx/Cbfa/AML factors.
TLDR
The biological functions of Runx factors in promoting cell fate determination and lineage progression are discussed, which include integrating ECM signaling and cues from developmental, hormonal, and signal transduction pathways by formation of complexes organized in subnuclear domains and mediating cell growth control.

TLDR
New concepts will be discussed within the context of understanding cellular responses of bone tissue that are supported by cooperative, synergistic and/or antagonistic activities at multiple basal and enhancer or suppressor sequences in gene promoters.
