Oxidative metabolism of cinnarizine (CZ) and its fluorine derivative flunarizine (FZ), both of which are selective calcium entry blockers, was examined in human liver microsomes. The ring-hydroxylations and the N-desalkylations constituted primary metabolic pathways in microsomal metabolism of CZ and FZ. Among these pathways, the ring-hydroxylase (p-hydroxylation) activities at the cinnamyl moiety of both drugs were highly correlated with debrisoquine 4-hydroxylase activity and CYP2D6 content…
Identification of the rat liver cytochrome P450 enzymes involved in the metabolism of the calcium channel blocker dipfluzine hydrochloride.
TLDR
This study aimed to identify the specific cytochrome P450 (CYP450) enzymes involved in the metabolism of dipfluzine hydrochloride using the combination of a chemical inhibition study, a correlation analysis and a panel of recombinant rat CYP450 enzymes to help understand drug-drug interactions when Dip is coadministered with other drugs.
Impact of P450 genetic polymorphism on the first-pass extraction of cardiovascular and neuroactive drugs.
This review highlights the present knowledge on the CYP2D6 (sparteine/debrisoquine) and the CYP2C19 (mephenytoin) polymorphisms. The relevant mutations at genomic level affecting protein expression
Cytochrome P450 Enzymes
The cytochrome P450 (CYP) enzymes, also known as the microsomal mixed function oxidase system, are the predominant biotransformation pathway in the body for lipid-soluble endogenous and xenobiotic
Isolation and characterization of metabolites of centpropazine in rat liver, intestine, and red blood cell homogenates.
TLDR
The potential sites for metabolism of centpropazine (CPZ) (an antidepressant) were evaluated in male Sprague-Dawley rats and the in vitro metabolic pathways for CPZ were proposed.
Cytochromes P450: A Structure-Based Summary of Biotransformations Using Representative Substrates
TLDR
A structure-based outline of CYP-catalyzed biotransformations of selected substrates, representing diverse structural classes of chemicals, ranging from drugs to toxicants is presented.
Polymorphism of Human Cytochrome P450 2D6 and Its Clinical Significance
TLDR
There is a considerable variability in the CYP2D6 allele distribution among different ethnic groups, resulting in variable percentages of PMs, IMs, EMs and UMs in a given population and the number of alleles is still growing.
Polymorphism of Human Cytochrome P450 2D6 and Its Clinical Significance
TLDR
CYP2D6 phenotyping and genotyping appear to be useful in predicting steady-state concentrations of some classical antipsychotic drugs, but their usefulness in predicting clinical effects must be explored.
Species difference in enantioselectivity for the oxidation of propranolol by cytochrome P450 2D enzymes.
TLDR
The results indicate that different properties of P450s belonging to the 2D subfamily cause the reversed enantioselectivity between human and monkey liver microsomes.
Possible pharmacokinetic and pharmacodynamic factors affecting parkinsonism inducement by cinnarizine and flunarizine.
TLDR
Some important roles are suggested of C-2 and F-2 in the development of parkinsonism as active metabolites during chronic medication with CZ and FZ, respectively.
"It's the genes, stupid". Molecular bases and clinical consequences of genetic cytochrome P450 2D6 polymorphism.
TLDR
This review highlights the information available on the genetic polymorphism of cytochrome P4502D6 expression in man and several lines of evidence indicate that CYP2D 6 could be involved in biotransformation of endogenous compounds.
