Soluble amyloid &bgr; peptide (A&bgr;) is believed to cause synaptic dysfunction in the early stages of Alzheimer's disease. Here, we examined in-vivo synaptic functions in the hippocampus in two lines of transgenic mice expressing different amounts of human wild-type amyloid precursor protein (APP). Compared with nontransgenic littermates, one transgenic line with higher APP expression displayed potent inhibition of paired-pulse facilitation and long-term potentiation in the absence of amyloid…

It is suggested that APP transgenic mice are a useful model of A β oligomer-induced pathology in the absence of amyloid plaques and that Aβ oligomers cause not only synaptic alteration but also other features of AD pathology.

A distortion of adult neurogenesis that is associated with Aβ production very early in amyloid pathogenesis is suggested and could prove useful as novel therapeutic targets and/or as early biomarkers of AD.

- BiologyReviews in the neurosciences

Transgenic (Tg) mouse models of Alzheimer’s disease are used to investigate mechanisms underlying disease pathology and identify therapeutic strategies, and common synaptic alterations in the various models that might underlie the memory deficits observed in these mice were searched for.

The data indicate that amyloid-beta deposits do not negatively impact plasticity in the brain and that overexpression of AbetaPP is not beneficial, but that normal estrogen levels are beneficial for plasticity.

It is concluded that APP may be cleaved via the general metabolic mechanisms of increased α- and γ-secretase levels, and that hyperphosphorylation of tau could be mediated by elevated levels of t Tau protein kinase, specifically in the precuneus and occipital cortex.
Intraneuronal amyloid β oligomers cause cell death via endoplasmic reticulum stress, endosomal/lysosomal leakage, and mitochondrial dysfunction in vivo

The findings suggest that intraneuronal Aβ oligomers cause cell death by inducing ER stress, endosomal/lysosomal leakage, and mitochondrial dysfunction in vivo.
APP Osaka Mutation in Familial Alzheimer’s Disease—Its Discovery, Phenotypes, and Mechanism of Recessive Inheritance
- BiologyInternational journal of molecular sciences
- 2020
The story behind the discovery of the Osaka mutation, the mutant’s phenotypes, and a mechanism of its recessive inheritance are described, which demonstrated that all pathologies of Alzheimer's disease can be induced by Aβ oligomers alone.
The E693Delta mutation in amyloid precursor protein increases intracellular accumulation of amyloid beta oligomers and causes endoplasmic reticulum stress-induced apoptosis in cultured cells.
- BiologyThe American journal of pathology
- 2009
The results suggest that mutant Abeta induces alteration of Abeta trafficking and subsequent ER stress-induced apoptosis via enhancement of its intracellular oligomerization, and suggest that Abeta oligomers exhibit toxicity in the extracellular space and within the cells themselves.
Regulation of cholesterol efflux by amyloid β secretion
- Biology, ChemistryJournal of neuroscience research

It is found that, when injected into mouse cerebral ventricle, reconstituted lipoproteins with Aβ were excreted into the peripheral tissues more efficiently than those without Aβ, which suggests that Aβ mediates cholesterol transport from the brain to the circulation.

- Biology, ChemistryThe Journal of Neuroscience

It is concluded that Aβ is synaptotoxic even in the absence of plaques and that high levels of Aβ1–42 are insufficient to induce plaque formation in mice expressing wild-type hAPP, supporting the emerging view that plaque-independent Aβ toxicity plays an important role in the development of synaptic deficits in AD and related conditions.

The data suggest that soluble non-fibrillar amyloid may contribute to the pathogenesis of AD both by impairing LTP/memory formation at the cellular level and by creating 'neuroplasticity imbalance' manifested by unopposed LTD in the setting of impaired capacity for neural repair via reversal of LTD or LTP.
