The T-helper-cell 1 and 2 (TH1 and TH2) pathways, defined by cytokines interferon-γ (IFN-γ) and interleukin-4 (IL-4), respectively, comprise two alternative CD4+ T-cell fates, with functional consequences for the host immune system. These cytokine genes are encoded on different chromosomes. The recently described TH2 locus control region (LCR) coordinately regulates the TH2 cytokine genes by participating in a complex between the LCR and promoters of the cytokine genes Il4, Il5 and Il13… Expand
Oct-1 regulates IL-17 expression by directing interchromosomal associations in conjunction with CTCF in T cells.
TLDR
Collectively, the data indicate Th17 lineage differentiation is restrained by the Th2 locus via interchromosomal associations organized by Oct-1 and CTCF. Expand
Activation-dependent intrachromosomal interactions formed by the TNF gene promoter and two distal enhancers
TLDR
TNF gene regulation reveals a mode of intrachromosomal interaction that combines a looped gene topology with interactions between enhancers and a gene promoter. Expand
Unconventional Association of the Polycomb Group Proteins with Cytokine Genes in Differentiated T Helper Cells*
TLDR
It is found that the expression of the polycomb group proteins, whose role in maintaining gene silencing is well documented, was induced during development in both T helper lineages, and their binding pattern at the cytokine genes was found to be a differential pattern. Expand
Distal Enhancer Elements Transcribe Intergenic RNA in the IL-10 Family Gene Cluster1
TLDR
It is speculated that these novel regulatory elements in the IL-10 family gene locus function via an intermediate regulatory RNA, which could account for the expression of these transcripts. Expand
Transvection-like interchromosomal interaction is not observed at the transcriptional level when tested in the Rosa26 locus in mouse
Long-range associations between enhancers and their target gene promoters have been shown to play critical roles in executing genome function. Recent variations of chromosome capture technology haveExpand
A distal conserved sequence element controls Ifng gene expression by T cells and NK cells.
TLDR
A single distal element may be essential for Ifng gene expression by both innate and adaptive immune effector cell lineages by using cross-species comparative sequence analyses. Expand
T helper cell differentiation: regulation by cis elements and epigenetics.
TLDR
Findings on the role of regulatory elements and their mechanisms of action are summarized, which are of broad significance for gene regulation, not only of the immune system but also of many, if not all, coregulated genes. Expand
Role of OCT-1 and partner proteins in T cell differentiation.
TLDR
The role of OCT-1 is described as a critical role in CD4 T cell differentiation and how this factor orchestrates the fate and function of CD4 effector T cells is discussed. Expand
Signal transduction pathways and transcriptional regulation in the control of Th17 differentiation.
The discovery of a new lineage of helper T cells that selectively produces interleukin (IL)-17 has provided exciting new insights into immunoregulation, host defense and the pathogenesis ofExpand
Virus Infection Induces NF-κB-Dependent Interchromosomal Associations Mediating Monoallelic IFN-β Gene Expression
TLDR
The stochastic and monoallelic expression of the IFN-beta gene depends on interchromosomal associations with three identified distinct genetic loci that could mediate binding of the limiting transcription factor NF-kappaB to the IFn-beta enhancer, thus triggering enhanceosome assembly and activation of transcription from this allele. Long-range intrachromosomal interactions in the T helper type 2 cytokine locus
TLDR
Using the chromosome conformation capture technique, it is found that in T cells, natural killer cells, B cells and fibroblasts, the promoters for the genes encoding TH2 cytokines are located in close spatial proximity, forming an initial chromatin core configuration. Expand
Th2-specific DNase I-hypersensitive sites in the murine IL-13 and IL-4 intergenic region.
TLDR
The correlation between differentiation into Th2 subtype and the appearance of HSS1 and HSS2 suggests that these regions may play a role in subtype-specific expression of the IL-13/IL-4 locus. Expand
Regulation of the Th2 cytokine locus by a locus control region.
TLDR
Investigation of the global locus-specific regulation of these genes using BAC transgenic mice containing the murine Th2 cytokine cluster carrying an IL4 promoter-luciferase reporter suggested the presence of an LCR in the locus. Expand
Hypersensitive site 7 of the TH2 locus control region is essential for expressing TH2 cytokine genes and for long-range intrachromosomal interactions
TLDR
It is shown that RHS7 is essential for the proper regulation of TH2 cytokine gene expression and did reduce long-range intrachromosomal interactions between the locus control region and promoters of the TH1 cytokine genes. Expand
Th2-specific chromatin remodeling and enhancer activity in the Th2 cytokine locus control region.
TLDR
This study identifies several functional elements within a 3' region of the rad50 gene possessing strong enhancer activity as well as activity consistent with function as a locus control region (LCR) for the flanking Th2 cytokine genes, and finds four DNase I hypersensitive clusters. Expand
Evolutionarily conserved sequence elements that positively regulate IFN-γ expression in T cells
TLDR
Two evolutionarily conserved noncoding sequence elements (IFNgCNS1 and IFNg CNS2) located upstream and downstream of the initiation codon of the murine Ifng gene are identified as two distal regulatory elements associated with T cell subset-specific IFN-γ expression. Expand
A Distal Enhancer in the Interferon-γ (IFN-γ) Locus Revealed by Genome Sequence Comparison*
TLDR
The approach as a successful method to detect cis-regulatory elements in the interferon-gamma (IFN-γ) gene is validated and combined with wet-lab techniques to illustrate a general and systematic method of identifying functional conserved regulatory regions of genes. Expand
Association of Transcriptionally Silent Genes with Ikaros Complexes at Centromeric Heterochromatin
TLDR
It is shown that transcriptionally inactive but not transcriptionally active genes associate with Ikaros-heterochromatin foci, which support a model of organization of the nucleus in which repressed genes are selectively recruited into centromeric domains. Expand
Cell-type-restricted binding of the transcription factor NFAT to a distal IL-4 enhancer in vivo.
