Rat hepatocyte cultures were incubated with [14C]dimethylnitrosamine (14C-NDMA) and the utility of measuring covalent binding of the radiolabel to newly synthesized albumin as a dose monitor of in vivo alkylation was determined. Hydrolysis of albumin, followed by derivatization and high performance liquid chromatography (HPLC) analysis of its constituent amino acids, showed radioactivity to be associated with a number of peaks.
