In a prospective-longitudinal study of a representative birth cohort, we tested why stressful experiences lead to depression in some people but not in others. A functional polymorphism in the promoter region of the serotonin transporter (5-HT T) gene was found to moderate the influence of stressful life events on depression. Individuals with one or two copies of the short allele of the 5-HT T promoter polymorphism exhibited more depressive symptoms, diagnosable depression, and suicidality in… Expand
5-HTTLPR genotype, stressful life events and late-life depression: No evidence of interaction in a French population
TLDR
In the only analysis known to the authors to be carried out in an exclusively elderly, European population, this null finding remained consistent after restriction of analysis to prevalent or incident cases. Expand
The interaction of stressful life events and a serotonin transporter polymorphism in the prediction of episodes of major depression: a replication.
TLDR
Variation at the 5-HTT moderates the sensitivity of individuals to the depressogenic effects of SLEs largely by producing, in SS individuals, an increased sensitivity to the impact of mild stressors. Expand
Interaction between genetic polymorphisms and stressful life events in first episode depression.
TLDR
Polymorphisms in the genes encoding the serotonin transporter and the brain derived neurotrophic factor interact with recent stressful life events on depression among patients with no history of previous depressive episodes. Expand
Interaction of Serotonin Transporter Gene-Linked Polymorphic Region and Stressful Life Events Predicts Cortisol Stress Response
TLDR
There was a significant interaction of 5-HTTLPR genotype and SLEs, but this interaction was only significant in younger adults and only when the measured SLES had occurred during the first 5 years of life, suggesting that both age and the specific type of SLE has a role in whether a significant gene–environment interaction is observed. Expand
An interaction between perceived stress and 5HTTLPR genotype in the prediction of stable depressive symptomatology.
TLDR
Analysis of data from the National Longitudinal Study of Adolescent Health (Add Health) indicates that the association between perceived stress and depression is moderated by 5HTTLPR genotype for females, but not for males. Expand
Stressful life events and the serotonin transporter gene (5-HTT) in recurrent clinical depression.
TLDR
This study failed to find evidence of gene-environment interplay in recurrent clinical depression, with no evidence of interaction between total SLEs and any of the 5-HTTLPR genetic models. Expand
HPA Axis Reactivity: A Mechanism Underlying the Associations Among 5-HTTLPR, Stress, and Depression
TLDR
These findings indicate that the 5-HTTLPR polymorphism is associated with biological stress reactivity, which may increase susceptibility to depression in the face of stressful life events. Expand
Suppression mediates the effect of 5-HTTLPR by stress interaction on depression.
TLDR
It is shown that individuals homozygous for the long allele respond to stressful events by reappraising their emotional meaning, which may hamper the harmful effect of stress on mental health. Expand
Development of depression: sex and the interaction between environment and a promoter polymorphism of the serotonin transporter gene.
TLDR
The results suggest that both the molecular and the psychosocial mechanisms underlying depression may differ between boys and girls and that males seem to be protected from depression. Expand
Gene-Environment Interactions in Depression Research
Recent studies on life stress, depression, and polymorphisms in the promoter region of the serotonin transporter gene (5-HTTLPR) are characterized by powerful genetic techniques, but are alsoExpand

Stressful life events and genetic liability to major depression: genetic control of exposure to the environment?
TLDR

Causal relationship between stressful life events and the onset of major depression.
TLDR
Stressed life events have a substantial causal relationship with the onset of episodes of major depression, however, about one-third of the association between stressful life events and onsets of depression is noncausal, since individuals predisposed to major depression select themselves into high-risk environments. Serotonin Transporter Genetic Variation and the Response of the Human Amygdala
TLDR
Genetically driven variation in the response of brain regions underlying human emotional behavior is demonstrated and differential excitability of the amygdala to emotional stimuli may contribute to the increased fear and anxiety typically associated with the short SLC6A4 allele. Expand

Diathesis-stress theories in the context of life stress research: implications for the depressive disorders.
TLDR
Advances in the conceptualization and measurement of life stress in the past 2 decades raise several questions concerning traditional diathesis-stress theories of psychopathology, and information is available on diatheses and stress for specific disorders to provide a foundation for more empirically based hypotheses about diathetic-stress interactions. Role of Genotype in the Cycle of Violence in Maltreated Children

Genetic perspectives on the serotonin transporter
TLDR
The combination of these comparative human and mouse studies may extend the opportunities to examine genetic alterations from a novel "bottom-up" approach [gene knockout or partial gene knockout in a combinational gene x gene x (yet unknown) gene approach], which is complementary to the traditional "top-down" genetic approach based upon studies of individuals with diagnosed neuropsychiatric disorders and their family members.
