This work reports the detailed molecular function of TGBp1, a suppressor of RNA silencing encoded by a potexvirus. TGBp1 interacts with SGS3 and RDR6 and aggregates SGS3/RDR6 bodies in the cytoplasm, thereby inhibiting dsRNA synthesis. Thus, this work sheds new light on the dsRNA synthesis–mediated secondary siRNA pathway as another general target of viral suppressors of RNA silencing. RNA silencing plays an important antiviral role in plants and invertebrates. To counteract antiviral RNA…

The Potyvirus Silencing Suppressor Protein VPg Mediates Degradation of SGS3 via Ubiquitination and Autophagy Pathways
TLDR
It is found that the VPg protein of Turnip mosaic virus (TuMV), which is a potyvirus, interacts with a host protein named SGS3, a key protein in the RNA silencing pathway.
Silencing suppressor protein VPg of a potyvirus interacts with the plant silencing-related protein SGS3.
TLDR
The data support the idea that VPg acts as a suppressor of RNA silencing and suggest that an interaction with SGS3 may be important, especially in suppression of sense-mediatedRNA silencing.
Viral RNase3 Co-Localizes and Interacts with the Antiviral Defense Protein SGS3 in Plant Cells
TLDR
Additional mechanisms needed for efficient RNase3-mediated suppression of RNAi are suggested and new information is provided about the subcellular context and phase of the RNAi pathway in whichRNase3 realizes RNAi suppression.
Viral RNase 3 Co-Localizes and Interacts with the Antiviral Defense Protein SGS 3 in Plant Cells
TLDR
Additional mechanisms needed for efficient RNase3-mediated suppression of RNAi are suggested and new information is provided about the subcellular context and phase of the RNAi pathway in whichRNase3 realizes RNAi suppression.
Geminivirus Activates ASYMMETRIC LEAVES 2 to Accelerate Cytoplasmic DCP2-Mediated mRNA Turnover and Weakens RNA Silencing in Arabidopsis
TLDR
AS2 is identified as a new component of plant P body complex which mediates mRNA decapping and degradation and promotes DCP2 decapping activity, accelerates mRNA turnover rate, inhibits siRNA accumulation and functions as an endogenous suppressor of PTGS.
Antiviral Defense Involves AGO4 in an Arabidopsis-Potexvirus Interaction.
TLDR
It is shown that AGO4 is also required for defense against the potexvirus Plantago asiatica mosaic virus (PlAMV), most likely independent of RdDM components such as DCL3, Pol IV, and Pol V.
Functional and Genetic Analysis Identify a Role for Arabidopsis ARGONAUTE5 in Antiviral RNA Silencing
TLDR
It is shown that all Arabidopsis AGO proteins have the potential to target PVX lacking its viral suppressor of RNA silencing (VSR), P25, but that only AGO2 and AGO5 are able to target wild-type PVX.
Antiviral Silencing and Suppression of Gene Silencing in Plants
TLDR
The current status of knowledge about the viral counter-defense strategies acting at various steps during virus infection in plants with the focus on representative, well studied silencing suppressor proteins is summarized.

Double-Stranded RNA Binding May Be a General Plant RNA Viral Strategy To Suppress RNA Silencing
TLDR
Results show that many plant viral silencing suppressors bind dsRNAs, suggesting that dsRNA binding is a general silencing suppression strategy which has evolved independently many times.
SDE5, the putative homologue of a human mRNA export factor, is required for transgene silencing and accumulation of trans-acting endogenous siRNA.
TLDR
A novel gene (SDE5) is reported here a novel gene that is required for transgene silencing and the production of trans-acting short interfering RNAs (tasiRNAs), and it is proposed that SDE5 acts together with RDR6 in generating double-stranded RNA from specific single- Stranded RNAs.
Arabidopsis RNA-Dependent RNA Polymerases and Dicer-Like Proteins in Antiviral Defense and Small Interfering RNA Biogenesis during Turnip Mosaic Virus Infection[W][OA]
TLDR
Systematic analysis of susceptibility and small RNA formation in Arabidopsis mutants lacking combinations of RNA-dependent RNA polymerase (RDR) and DCL proteins revealed that the vast majority of virus-derived small interfering RNAs (siRNAs) were dependent on DCL4 and RDR1, although full antiviral defense also required DCL2 and R DR6.
Variability in the level of RNA silencing suppression caused by triple gene block protein 1 (TGBp1) from various potexviruses during infection.
TLDR
It is suggested that suppressor activities differ even among homologous proteins encoded by viruses of the same genus, and that TGBp1 contributes to the variation in the level of RNA silencing suppression by potexviruses.
Transitivity in Arabidopsis can be primed, requires the redundant action of the antiviral Dicer-like 4 and Dicer-like 2, and is compromised by viral-encoded suppressor proteins.
TLDR
How several viral silencing suppressors inhibit the RDR6-dependent amplification of virus-induced and transgene-induced gene silencing is reported, and in vivo evidence supporting a key role for priming in this specific reaction is provided.

TLDR
A sophisticated viral counter-defense strategy that targets the silencing antiviral pathway at multiple steps is revealed and may be essential for protecting CTV with such a large RNA genome from antiviral silencing in the perennial tree host.
Cucumber mosaic virus-encoded 2b suppressor inhibits Arabidopsis Argonaute1 cleavage activity to counter plant defense.
TLDR
It is concluded that 2b blocks AGO1 cleavage activity to inhibit miRNA pathways, attenuate RNA silencing, and counter host defense, providing insight on the molecular arms race between host antiviral RNAsilencing and virus counterdefense.
Viral Protein Inhibits RISC Activity by Argonaute Binding through Conserved WG/GW Motifs
TLDR
It is demonstrated that in Sweet potato mild mottle virus (SPMMV), the role of silencing suppressor is played by the P1 protein despite the presence in its genome of an HC-Pro protein, which, in potyviruses, acts as the suppressor.
Recognition of small interfering RNA by a viral suppressor of RNA silencing
TLDR
The 1.85-Å crystal structure of p19 bound to a 21-nucleotide siRNA is reported, where the 19-base-pair RNA duplex is cradled within the concave face of a continuous eight-stranded β-sheet, formed across the p19 homodimer interface.
An RNA-Dependent RNA Polymerase Prevents Meristem Invasion by Potato Virus X and Is Required for the Activity But Not the Production of a Systemic Silencing Signal
TLDR
A model of virus defense in which RDR6 uses incoming silencing signal to generate double-stranded RNA precursors of secondary siRNA mediate RNA silencing as an immediate response that slows down the systemic spreading of the virus into the growing point and newly emerging leaves is suggested.
