This work reports the detailed molecular function of TGBp1, a suppressor of RNA silencing encoded by a potexvirus. TGBp1 interacts with SGS3 and RDR6 and aggregates SGS3/RDR6 bodies in the cytoplasm, thereby inhibiting dsRNA synthesis. Thus, this work sheds new light on the dsRNA synthesis–mediated secondary siRNA pathway as another general target of viral suppressors of RNA silencing.
