ABSTRACT The HESI-led RISK21 effort has developed a framework supporting the use of twenty-first century technology in obtaining and using information for chemical risk assessment. This framework represents a problem formulation-based, exposure-driven, tiered data acquisition approach that leads to an informed decision on human health safety to be made when sufficient evidence is available. It provides a transparent and consistent approach to evaluate information in order to maximize the…

Use of the RISK21 roadmap and matrix: human health risk assessment of the use of a pyrethroid in bed netting

The evaluation demonstrated the concept of using existing information within the RISK21 matrix to drive the generation of additional data using a value-of-information approach and showed how new methodology such as the use of in vitro studies and assays could be used to answer the specific questions which arise through the useof the matrix.

Prioritization of chemicals in food for risk assessment by integrating exposure estimates and new approach methodologies: A next generation risk assessment case study

Next generation risk assessment is defined as a knowledge-driven system that allows for cost-efficient assessment of human health risk related to chemical exposure, without animal experimentation.

Setting the stage for next-generation risk assessment with non-animal approaches: the EU-ToxRisk project experience

The EU-ToxRisk consortium has collated its scientific and regulatory learnings from this particular project objective to implement systems toxicology-guided, animal-free next-generation risk assessment.

Establishing a quantitative framework for regulatory interpretation of genetic toxicity dose–response data: Margin of exposure case study of 48 compounds with both in vivo mutagenicity and carcinogenicity dose–response data

The results indicate that regulatory decisions based on in vivo genotoxicity dose– response data would be consistent with those based on carcinogenicity dose–response data; in some cases, genot toxicity‐based decisions would be more conservative.

Letter to the editor RE: Wolf et al.1

This letter further refines the exposure estimates for TDI based on additional insight into the physical–chemical properties, as well as publicly available and reliable information, and uses the Threshold of Toxicological Concern approach embodied in the RISK21 Roadmap to demonstrate a more appropriate assessment of human health risk.

Risk assessment in the 21st century: Roadmap and matrix

Abstract The RISK21 integrated evaluation strategy is a problem formulation-based exposure-driven risk assessment roadmap that takes advantage of existing information to graphically represent the

A 21st century roadmap for human health risk assessment

This paper provides a brief overview of the tiered RISK21 framework called the roadmap and risk visualization matrix, and articulates the core principles derived by RISk21 participants that guided its development.

Risk assessment of combined exposure to multiple chemicals: A WHO/IPCS framework.

An evaluation of the implementation of the Cramer classification scheme in the Toxtree software

Overall Toxtree was found to be a useful tool in facilitating the systematic evaluation of compounds through the Cramer scheme and a number of inconsistencies were examined in turn and rationalised as far as possible.

Science and Decisions: Advancing Risk Assessment

The NRC report tackles a number of topics relating to improvements in the process, including the design and framing of risk assessments, uncertainty and variability characterization, selection and use of defaults, unification of cancer and noncancer dose-response assessment, cumulative risk assessment, and the need to increase EPA's capacity to address these improvements.
