The structural organization of nongastric H-K-ATPase, unlike that of closely related Na-K-ATPase and gastric H-K-ATPase, is not well characterized. Recently, we demonstrated that nongastric H-K-ATPase alpha-subunit (alpha(ng)) is expressed in apical membranes of rodent prostate. Its highest level, as well as relative abundance, with respect to alpha(1)-isoform of Na-K-ATPase, was observed in anterior lobe. Here, we aimed to determine the subunit composition of nongastric H-K-ATPase through the…

Loss of acidification of anterior prostate fluids in Atp12a-null mutant mice indicates that nongastric H-K-ATPase functions as proton pump in vivo.
TLDR
Results show that nongastric H-K-ATPase is required for acidification of luminal prostate fluids, thereby providing a strong in vivo correlate of previous functional expression studies demonstrating that it operates as a proton pump.
Pharmacological profiles of the murine gastric and colonic H,K-ATPases.
Polarized membrane distribution of potassium-dependent ion pumps in epithelial cells: Different roles of the N-glycans of their β subunits
TLDR
The Na,K-ATPase β1-subunit is the major β-sub unit isoform in cells with lateral location of the pump and all three N-glycans are important for the lateral membrane retention of the Pump due to glycan-mediated interaction between the β 1-subunits of the two neighboring cells in the cell monolayer and cytosolic linkage of the α- subunit to the cytoskeleton.
An Extracellular Loop of the Human Non-Gastric H,K-ATPase a-subunit is Involved in Apical Plasma Membrane Polarization
TLDR
This work shows in transfected Madin-Darby canine kidney (MDCK) cells that the related non-gastric H,KATPase, ATP1AL1, does contain similar sorting motifs in close proximity to TM4, providing further evidence that the cellular distribution of P-type ATPases is determined by conformational sorted motifs.
The Non-Gastric H+/K+ ATPase (ATP12A) Is Expressed in Mammalian Spermatozoa
TLDR
Results are consistent with those observed for the β1 subunit of Na+/K+ ATPase, suggesting that the latter may assemble with the α subunit to produce a functional ATP12A dimer in sperm cells, and hypothesize that ATP 12A may play a role in acrosome reactions.

TLDR
The findings discussed in this review suggest that, as in the famous story, the ugly duckling of the X-K-ATPase family is actually a swan.
Role of homologous ASP334 and GLU319 in human non-gastric H,K- and Na,K-ATPases in cardiac glycoside binding.
Enzymatic Activity of Renal H-K-ATPase in the Outer Medullary Collecting Duct of Transgenic Mice
TLDR
The results demonstrated potassium depletion’s association with increased EA of H-K-ATPase in OMCD, consistent with the activation of HKA α2 isoform, and a significant difference in ATP production in HKAα2 KO mice is likely due to enhanced EA ofH- ATPase under potassium depletion.
The renal H+-K+-ATPases: physiology, regulation, and structure.
TLDR
Evaluation of enzymatic functions along the nephron and collecting duct and studies in HKalpha(1) andHKalpha(2) knockout mice suggest that the H(+)-K( +)-ATPases may function to transport ions other than protons and potassium.
The H+- and H+, K+-ATPases of the Collecting Duct
TLDR
The goal of this chapter is to consider the molecular properties of the plasma membrane H+-ATPase and the H+,K+- ATPases of the collecting duct.

TLDR
Tissue-specific upregulation of this β-subunit mRNA in response to K depletion, localization of its protein, its upregulation by K depletion in apical membranes of distal colon, and its physical association with HKcα protein provide compelling evidence that HKcβ is the putative β- subunit of colonic H-K-ATPase.
