}The structural organization of nongastric H-K-ATPase, unlike that of closely related Na-K-ATPase and gastric H-K-ATPase, is not well characterized. Recently, we demonstrated that nongastric H-K-ATPase alpha-subunit (alpha(ng)) is expressed in apical membranes of rodent prostate. Its highest level, as well as relative abundance, with respect to alpha(1)-isoform of Na-K-ATPase, was observed in anterior lobe. Here, we aimed to determine the subunit composition of nongastric H-K-ATPase through the…

Results show that nongastric H-K-ATPase is required for acidification of luminal prostate fluids, thereby providing a strong in vivo correlate of previous functional expression studies demonstrating that it operates as a proton pump.

The Na,K-ATPase β1-subunit is the major β-sub unit isoform in cells with lateral location of the pump and all three N-glycans are important for the lateral membrane retention of the Pump due to glycan-mediated interaction between the β 1-subunits of the two neighboring cells in the cell monolayer and cytosolic linkage of the α- subunit to the cytoskeleton.

This work shows in transfected Madin-Darby canine kidney (MDCK) cells that the related non-gastric H,KATPase, ATP1AL1, does contain similar sorting motifs in close proximity to TM4, providing further evidence that the cellular distribution of P-type ATPases is determined by conformational sorted motifs.

Results are consistent with those observed for the β1 subunit of Na+/K+ ATPase, suggesting that the latter may assemble with the α subunit to produce a functional ATP12A dimer in sperm cells, and hypothesize that ATP 12A may play a role in acrosome reactions.

The results demonstrated potassium depletion’s association with increased EA of H-K-ATPase in OMCD, consistent with the activation of HKA α2 isoform, and a significant difference in ATP production in HKAα2 KO mice is likely due to enhanced EA ofH- ATPase under potassium depletion.
