The assessment of glycosyl-scanning to expand the molecular and functional diversity of metabolites from the underground coal mine fire-associated Streptomyces sp. RM-14-6 is reported. Using the engineered glycosyltransferase OleD Loki and a 2-chloro-4-nitrophenylglycoside-based screen, six metabolites were identified as substrates of OleD Loki, from which 12 corresponding metabolite glycosides were produced and characterized. This study highlights the first application of the 2-chloro-4…

OleD Loki as a Catalyst for Hydroxamate Glycosylation
Herein we describe the ability of the permissive glycosyltransferase (GT) OleD Loki to convert a diverse set of >15 histone deacetylase (HDAC) inhibitors (HDACis) into their corresponding hydroxamate
Sugar-pirating as an enabling platform for the synthesis of 4,6-dideoxyhexoses.
An efficient divergent synthetic strategy that leverages the natural product spectinomycin to access uniquely functional-ized monosaccharides is described and applied to the synthesis of two function-alized sugars commonly associated with macrolide antibiotics.
Structural Reassignment and Absolute Stereochemistry of Madurastatin C1 (MBJ-0034) and the Related Aziridine Siderophores: Madurastatins A1, B1, and MBJ-0035.
NMR spectroscopy in combination with partial synthesis allowed confirmation of the structure of madurastatin C1 (1) as containing an N-terminal 2-(2-hydroxyphenyl)oxazoline in place of the originally postulated aziridine, while absolute stereochemistry was determined via Harada's advanced Marfey's method.
Modulating the Serotonin Receptor Spectrum of Pulicatin Natural Products.
The results reveal the 2-arylthiazoline scaffold as a tunable serotonin receptor-targeting pharmacophore and tests in mice show potential antiseizure and antinociceptive activities at high doses without motor impairment.
A multi-tiered screening of a natural product library and validation process to provide novel information about the mechanisms underlying EtOH-induced changes in neurobiology and to identify novel chemical scaffolds to be exploited in the development of pharmacological treatments for AUD is undertaken.

Spoxazomicin D and Oxachelin C, Potent Neuroprotective Carboxamides from the Appalachian Coal Fire-Associated Isolate Streptomyces sp. RM-14-6.
Bioactivity analysis revealed 2 and 5 as potent neuroprotectives and lenoremycin (9) and its sodium salt (10) as potent UPR modulators, highlighting new functions for phenol-oxazolines/salicylates and polyether pharmacophores.

Probing the aglycon promiscuity of an engineered glycosyltransferase.
This study highlights the ability of OleD variants to glucosylate a total of 71 diverse acceptors, catalyze iterative glycosylation with numerous substrates, and establishes OleD as the first multifunctional GT capable of generating O-, S- and N-glycosides.
Cytotoxic Indolocarbazoles from Actinomadura melliaura ATCC 39691.
The slightly improved anticancer potency of the newly reported N-6-desmethyl 1 (compared to 6) contrasts extensive SAR of monoglycosylated rebeccamycin-type topoisomerase I inhibitors where N- 6 alkylation has contributed to improved potency and ADME.
Natural glycoconjugates with antitumor activity.
This review focuses on natural glycoconjugates with anticancer properties and describes anthracyclines, aureolic acids, enediyne antibiotics, macrolide and glycopeptides presenting different strengths and mechanisms of action against human cancers.
O-Glycosylation methods in the total synthesis of complex natural glycosides.
The total syntheses of 33 complex natural O-glycosides, such as the glycosides of macrocyclic lactones/lactams, enediynes, angucyclines, and anthracyclines, are highlighted, with a major focus being
