Cardiac fibroblasts (CFs) are an important cellular component of myocardial responses to injury and to hypertrophic stimuli. We studied G protein-coupled receptors to understand how CFs integrate signals that activate G(q), G(s), and G(i). We predicted that the second messenger pathways present in CFs were distinct from those in cardiac myocytes and that unique signaling interactions existed in the CFs. ANG II, bradykinin, ATP, and UTP stimulated inositol phosphate (IP) production 2.2- to 7…

Figures and Tables from this paper

Protein kinase C contributes to desensitization of ANG II signaling in adult rat cardiac fibroblasts.
TLDR
A large portion of the IP-Ca(2+) responses of rat CFs to ANG II are short-lived because of rapid, PKC-mediated desensitization, which is demonstrated to be proportional to G(q)-phospholipase C-IP(3) activity under the conditions studied.
Adenosine receptors and second messenger signaling pathways in rat cardiac fibroblasts.
TLDR
It is concluded that a combination of cAMP-dependent signals generated via A( 2a) and A(2b) receptors likely mediate ADO signaling in adult rat CF.
Pharmacology of G-Protein-linked Signaling in Cardiac Fibroblasts
TLDR
It is believed that activation and blockade of cell surface receptors, especially G-protein linked, could be fruitful way of manipulating CF function in vivo.
Angiotensin II Enhances Adenylyl Cyclase Signaling via Ca2+/Calmodulin
TLDR
Data indicate that ANG II potentiates cAMP formation via Ca2+-dependent activation of AC activity, which in turn attenuates collagen synthesis and demonstrates one functional consequence of cross-talk between Gq and Gs signaling pathways in cardiac fibroblasts.
Crosstalk Between the Renin–Angiotensin System and the Advance Glycation End Product Axis in the Heart: Role of the Cardiac Fibroblast
TLDR
Because cardiac fibrosis is an important component of diabetic cardiomyopathy, new data is included that suggests a possible crosstalk between the RAS and AGE/RAGE pathway in order to activate CFs in diabetes.
Cloning, expression, signaling mechanisms, and membrane targeting of P2Y(11) receptors in Madin Darby canine kidney cells.
TLDR
The data support the BL localization of cP2Y(11) receptors and receptor coupling to changes in I(sc) in MDCK-D1 cells except in cases in which receptors are overexpressed; receptor overexpression leads to altered sensitivities and sites of coupling to physiologic responses.
Phenylephrine promotes cardiac fibroblast proliferation through calcineurin-NFAT pathway.
TLDR
The results suggest that the Ca(2+)/CaN/NFAT pathway mediates PE-induced CF proliferation, and this pathway might be a possible therapeutic target in cardiac fibrosis.

Involvement of protein kianse C and Ca2+ in angiotensin II-induced mitogenesis of cardiac fibroblasts.
TLDR
Angiotensin (ANG) II can activate MAP kinase in cardiac fibroblasts by either Ca(2+)- or PKC-dependent pathways, and whereas the full effect of PDGF-BB on thymidine incorporation and cell proliferation requires a phorbol ester-sensitive PKC, the hyperplastic growth effect of ANG II does not.
Functional coupling of the beta 2-adrenoceptor to a pertussis toxin-sensitive G protein in cardiac myocytes.
TLDR
It is shown that pertussis toxin (PTX) pretreatment specifically potentiates the responses of rat heart cells to beta 2AR but not beta 1AR stimulation, and that the Gs-coupled beta 2 AR can simultaneously activate a pathway that leads to functional inhibition in cardiac cells via a PTX-sensitive G protein.
Role of AT1 and AT2 receptors in regulation of MAPKs and MKP-1 by ANG II in adult cardiac myocytes.
TLDR
There is constitutive as well as inducible suppression of ERKs and c-jun NH2-terminal protein kinases by MKP and PP-2A/PP-1 in the adult cardiac myocyte phenotype.
Coupling of the type A endothelin receptor to multiple responses in adult rat cardiac myocytes.
TLDR
The effects of ET on transmembrane signaling in rat ventricular myocytes result from occupation of ETA receptors and that the responses are likely to be long lived, compared with those of the readily dissociable neurotransmitters released by the autonomic nervous system.
Angiotensin II and other hypertrophic stimuli mediated by G protein-coupled receptors activate tyrosine kinase, mitogen-activated protein kinase, and 90-kD S6 kinase in cardiac myocytes. The critical role of Ca(2+)-dependent signaling.
TLDR
Results suggest that Ang II and other hypertrophic stimuli, known to act through Gq protein-coupled receptors, rapidly cause tyrosine phosphorylation of several intracellular substrates through activation of tyrosin kinase and activate MAP kinases and RSK in cardiac myocytes as well as in cardiac fibroblasts.
Endothelin ETA receptor regulates signaling and ANF gene expression via multiple G protein-linked pathways.
TLDR
Data indicate that the ETA receptor is the exclusive receptor on neonatal ventricular myocytes and that this receptor couples to both phosphoinositide hydrolysis and adenylyl cyclase.
Endothelin inhibits adenylate cyclase and stimulates phosphoinositide hydrolysis in adult cardiac myocytes.
Angiotensin II and Other Hypertrophic Stimuli Mediated by G Protein–Coupled Receptors Activate Tyrosine Kinase, Mitogen-Activated Protein Kinase, and 90-kD S6 Kinase in Cardiac Myocytes The Critical Role of Ca2+-Dependent Signaling
TLDR
Results suggest that Ang II and other hypertrophic stimuli, known to act through G q protein–coupled receptors, rapidly cause tyrosine phosphorylation of several intracellular substrates through activation of tyrosin kinase and activate MAP kinases and RSK in cardiac myocytes as well as in cardiac fibroblasts.
Quantification of signalling components and amplification in the beta-adrenergic-receptor-adenylate cyclase pathway in isolated adult rat ventricular myocytes.
TLDR
The data presented here demonstrate that, in a physiologically relevant setting, G-protein is present in large stoichiometric excess relative to both receptor and effector, and shows that adenylate cyclase is the component distal to receptor that limits agonist-mediated increases in cyclic AMP production.
Angiotensin II‐elicited signal transduction via AT1 receptors in endothelial cells
TLDR
The aim of this work was to characterize the AII receptor subtypes in rat aortic endothelial cells (RAEC) in primary culture and to evaluate the signal pathways coupled to these receptors by measuring the activation of PLC and phospholipase A2 (PLA2).
