To gain further insight into the genetic architecture of psoriasis, we conducted a meta-analysis of 3 genome-wide association studies (GWAS) and 2 independent data sets genotyped on the Immunochip, including 10,588 cases and 22,806 controls. We identified 15 new susceptibility loci, increasing to 36 the number associated with psoriasis in European individuals. We also identified, using conditional analyses, five independent signals within previously known loci. The newly identified loci shared…

Enhanced meta-analysis and replication studies identify five new psoriasis susceptibility loci

The combined analysis, consisting of over 15,000 cases and 27,000 controls, identifies five new psoriasis susceptibility loci at genomewide significance, and demonstrates that NFKBIZ is a TRAF3IP2–dependent target of IL-17 signaling in human skin keratinocytes, thereby functionally linking two strong candidate genes.

Dense genotyping of immune-related susceptibility loci reveals new insights into the genetics of psoriatic arthritis

Key insights into the genetics of PsA are identified that could begin to explain fundamental differences between psoriasis and PsA.

Candidate gene polymorphisms and risk of psoriasis: A pilot study

Six SNPs were associated with an increased risk of developing psoriasis in the present study, and Fisher's exact test detected statistical significance; however, following false discovery rate and Bonferroni correction, this association was no longer significant.

Large scale meta-analysis characterizes genetic architecture for common psoriasis associated variants

This study provides a comprehensive layout for the genetic architecture of common variants for psoriasis, including data from eight different Caucasian cohorts, with a combined effective sample size >39,000 individuals.

Identification of a missense variant in LNPEP that confers psoriasis risk.

A genetic susceptibility factor was identified and genetic evidence of insight into psoriasis pathogenesis with the involvement of the renin-angiotensin system pathway was provided, consistent with clinical observations.

Meta-analysis of shared genetic architecture across ten pediatric autoimmune diseases

Network and protein-interaction analyses demonstrated converging roles for the signaling pathways of type 1, 2 and 17 helper T cells (TH1, TH2 and TH17), JAK-STAT, interferon and interleukin in multiple autoimmune diseases.

Five regulatory genes detected by matching signatures of eQTL and GWAS in psoriasis.

Meta-analysis of shared genetic architecture across ten pediatric autoimmune diseases

An χ2 meta-analysis across ten pediatric-age-of-onset autoimmune diseases (pAIDs) in a casecontrol study suggests that genetic predisposition underlies disease susceptibility.

Exome-wide association study reveals novel psoriasis susceptibility locus at TNFSF15 and rare protective alleles in genes contributing to type I IFN signalling

Previous reports of protective low-frequency protein-altering variants within IFIH1 and TYK2 (encoding an innate antiviral receptor and Janus kinase) are validated, establishing a further series of protective rare variants.

A genome-wide association study identifies new psoriasis susceptibility loci and an interaction between HLA-C and ERAP1

These findings implicate pathways that integrate epidermal barrier dysfunction with innate and adaptive immune dysregulation in psoriasis pathogenesis and report compelling evidence for an interaction between the HLA-C and ERAP1 loci.

Insights into psoriasis and other inflammatory diseases from large-scale gene expression studies.

A more comprehensive analysis of the transcriptional changes that occur in involved and uninvolved skin of 15 psoriatic patients versus six normal controls showed differences independent of the gender, age, skin site and HLA class I status of the patient.

Genome-wide association study identifies a psoriasis susceptibility locus at TRAF3IP2

A genome-wide association analysis of 2,339,118 SNPs in 472 PsV cases and 1,146 controls from Germany suggests that TRAF3IP2 represents a shared susceptibility for PsV and PsA.

Meta-analysis of genome-wide association studies in celiac disease and rheumatoid arthritis identifies fourteen non-HLA shared loci

Background and objectives Epidemiology and candidate gene studies indicate a shared genetic basis for celiac disease (CD) and rheumatoid arthritis (RA), but the extent of this sharing has not been

An Immune Response Network Associated with Blood Lipid Levels

A gene network linking blood lipids and circulating cell types is uncovered and insight is offered into the hypothesis that the inflammatory response plays a prominent role in metabolism and the potential control of atherogenesis is offered.

Assessment of the psoriatic transcriptome in a large sample: additional regulated genes and comparisons with in vitro models.

This comprehensive analysis of differentially regulated transcripts in psoriasis provides additional insight into the pathogenic mechanisms involved and emphasizes the need for more complex yet tractable experimental models of psoriatic disease.

Association analyses identify six new psoriasis susceptibility loci in the Chinese population

Six new susceptibility loci associated with psoriasis in the Chinese study containing the candidate genes ERAP1, PTTG1, CSMD1, GJB2, SERPINB8 and ZNF816A are identified and the heterogeneity of disease susceptibility between the Chinese and European populations is highlighted.

Psoriasis genome-wide association study identifies susceptibility variants within LCE gene cluster at 1q21

This is the first large genome-wide association study in a Chinese population to identify susceptibility variants for psoriasis using a two-stage case-control design and identified a new susceptibility locus within the LCE gene cluster on 1q21.
