To gain further insight into the genetic architecture of psoriasis, we conducted a meta-analysis of 3 genome-wide association studies (GWAS) and 2 independent data sets genotyped on the Immunochip, including 10,588 cases and 22,806 controls. We identified 15 new susceptibility loci, increasing to 36 the number associated with psoriasis in European individuals. We also identified, using conditional analyses, five independent signals within previously known loci. The newly identified loci shared… Expand
Enhanced meta-analysis and replication studies identify five new psoriasis susceptibility loci
TLDR
The combined analysis, consisting of over 15,000 cases and 27,000 controls, identifies five new psoriasis susceptibility loci at genomewide significance, and demonstrates that NFKBIZ is a TRAF3IP2–dependent target of IL-17 signaling in human skin keratinocytes, thereby functionally linking two strong candidate genes. Expand
Dense genotyping of immune-related susceptibility loci reveals new insights into the genetics of psoriatic arthritis
TLDR
Key insights into the genetics of PsA are identified that could begin to explain fundamental differences between psoriasis and PsA. Expand
Candidate gene polymorphisms and risk of psoriasis: A pilot study
TLDR
Six SNPs were associated with an increased risk of developing psoriasis in the present study, and Fisher's exact test detected statistical significance; however, following false discovery rate and Bonferroni correction, this association was no longer significant. Expand
Large scale meta-analysis characterizes genetic architecture for common psoriasis associated variants
TLDR
This study provides a comprehensive layout for the genetic architecture of common variants for psoriasis, including data from eight different Caucasian cohorts, with a combined effective sample size >39,000 individuals. Expand
Identification of a missense variant in LNPEP that confers psoriasis risk.
TLDR
A genetic susceptibility factor was identified and genetic evidence of insight into psoriasis pathogenesis with the involvement of the renin-angiotensin system pathway was provided, consistent with clinical observations. Expand
Meta-analysis of shared genetic architecture across ten pediatric autoimmune diseases
TLDR
Network and protein-interaction analyses demonstrated converging roles for the signaling pathways of type 1, 2 and 17 helper T cells (TH1, TH2 and TH17), JAK-STAT, interferon and interleukin in multiple autoimmune diseases. Expand
Five regulatory genes detected by matching signatures of eQTL and GWAS in psoriasis.
TLDR
Five genes with implied regulatory effect were revealed to be associated significantly with the risk of psoriasis, with one novel signal in FAM20B gene which is significantly expressed (P=3.24×10(-5)). Expand
Meta-analysis of shared genetic architecture across ten pediatric autoimmune diseases
Genome-wide association studies (GWASs) have identified hundreds of susceptibility genes, including shared associations across clinically distinct autoimmune diseases. We performed an inverse χ2Expand
Exome-wide association study reveals novel psoriasis susceptibility locus at TNFSF15 and rare protective alleles in genes contributing to type I IFN signalling
TLDR
Previous reports of protective low-frequency protein-altering variants within IFIH1 and TYK2 (encoding an innate antiviral receptor and Janus kinase) are validated, establishing a further series of protective rare variants. Expand
Cellular dissection of psoriasis for transcriptome analyses and the post-GWAS era
BackgroundGenome-scale studies of psoriasis have been used to identify genes of potential relevance to disease mechanisms. For many identified genes, however, the cell type mediating disease activityExpand

A genome-wide association study identifies new psoriasis susceptibility loci and an interaction between HLA-C and ERAP1
TLDR
These findings implicate pathways that integrate epidermal barrier dysfunction with innate and adaptive immune dysregulation in psoriasis pathogenesis and report compelling evidence for an interaction between the HLA-C and ERAP1 loci. Expand
Insights into psoriasis and other inflammatory diseases from large-scale gene expression studies.
TLDR
A more comprehensive analysis of the transcriptional changes that occur in involved and uninvolved skin of 15 psoriatic patients versus six normal controls showed differences independent of the gender, age, skin site and HLA class I status of the patient. Expand
Multiple common variants for celiac disease influencing immune gene expression
TLDR
Variants from 13 new regions reached genome-wide significance and most contain genes with immune functions, with ETS1, RUNX3, THEMIS and TNFRSF14 having key roles in thymic T-cell selection. Expand
Molecular dissection of psoriasis: integrating genetics and biology.
TLDR
The epidemiology, immunopathology, and genetics of psoriasis are reviewed, and a disease model integrating its genetics and immunology is presented, using cases and controls rather than families. Expand
Genome-wide association study identifies a psoriasis susceptibility locus at TRAF3IP2
TLDR
A genome-wide association analysis of 2,339,118 SNPs in 472 PsV cases and 1,146 controls from Germany suggests that TRAF3IP2 represents a shared susceptibility for PsV and PsA. Expand
Meta-analysis of genome-wide association studies in celiac disease and rheumatoid arthritis identifies fourteen non-HLA shared loci
Background and objectives Epidemiology and candidate gene studies indicate a shared genetic basis for celiac disease (CD) and rheumatoid arthritis (RA), but the extent of this sharing has not beenExpand
An Immune Response Network Associated with Blood Lipid Levels
TLDR
A gene network linking blood lipids and circulating cell types is uncovered and insight is offered into the hypothesis that the inflammatory response plays a prominent role in metabolism and the potential control of atherogenesis is offered. Expand
Multiple Loci within the Major Histocompatibility Complex Confer Risk of Psoriasis
TLDR
It is demonstrated that there are at least two additional loci within the MHC conferring risk of psoriasis within the human leukocyte antigen (HLA) region, independently of HLA-Cw*0602 and the C6orf10 locus, suggesting the potential pathogenic involvement of Hla-B. Expand
Combined analysis of genome-wide association studies for Crohn disease and psoriasis identifies seven shared susceptibility loci.
TLDR
The usefulness of joint analyses of clinically distinct immune-mediated diseases and the map of shared genetic risk loci revealed in silico expression quantitative trait locus analyses revealed that the associations at ZMIZ1 and near SOCS1 have a potential functional effect on gene expression.
