Icariin is a major bioactive pharmaceutical constituent isolated from Chinese medicine Horny Goat Weed (Ying Yang Huo) with potent cardiovascular protective functions. Emerging evidence in the past decade has shown that Icariin possesses multiple atheroprotective functions, through multiple mechanisms, including attenuating DNA damage, correcting endothelial dysfunction, inhibiting the proliferation and migration of smooth muscle cells, repressing macrophage-derived foam cell formation and…

E. koreanum inhibited platelet aggregation and thrombus formation by attenuating calcium mobilization, ATP secretion, TXB2 formation, and integrin αIIbβ3 activation, and it may be considered as a potential candidate to treat and prevent platelet-related cardiovascular disorders.
Icariin, an Up-and-Coming Bioactive Compound Against Neurological Diseases: Network Pharmacology-Based Study and Literature Review
A protein–protein interaction network is constructed and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes functional enrichment analyses are carried out to predict the therapeutic actions of icariin in nervous system diseases including Alzheimer disease, Parkinson disease, ischemic stroke, depressive disorder, multiple sclerosis, glioblastoma, and hereditary spastic paraplegias.
Inhibition of airway remodeling and inflammatory response by Icariin in asthma
The fact that ICA can alleviate OVA-induced asthma at least partly through inhibition of ASMC proliferation via MAPK/Erk pathway provides a solid theoretical basis for ICA as a replacement therapy for asthma.
Icariin Attenuates High Glucose-Induced Apoptosis, Oxidative Stress, and Inflammation in Human Umbilical Venous Endothelial Cells.
It is found that high glucose markedly induced cell apoptosis, enhanced reactive oxygen species generation, and elevated expression levels of inflammatory factors and cell adhesion molecules, which were greatly subdued by icariin supplementation, which provides a promising strategy for future treatment of diabetic vascular complications.
Optimized Icariin Phytosomes Exhibit Enhanced Cytotoxicity and Apoptosis-Inducing Activities in Ovarian Cancer Cells
Phytosome formulation of ICA significantly potentiates its cytotoxic activities against OVCAR-3 cells, mediated, at least partly, by enhanced ICA cellular permeation, apoptosis, and ROS.

Atheroprotective Effects and Molecular Targets of Tanshinones Derived From Herbal Medicine Danshen
A new perspective for advancing the understanding of the “ancient” herb Danshen from “modern” biomedical perspectives is provided, supporting the possibility of exploiting tanshinones and derivatives as effective therapeutics against atherosclerosis‐related cardiovascular and metabolic diseases.
Effects and mechanisms of icariin on atherosclerosis.
Icariin can prevent atherosclerotic lesion and its mechanism may be that it can defend against the oxidation damage to HUVECs, inhibit the adhesion of monocyte to HUVCs, and reduce the secretion and expression of adhesion molecules including ICAM-1, VCAM- 1, E-selectin.
Icariin inhibits atherosclerosis progress in Apoe null mice by downregulating CX3CR1 in macrophage.
Cryptotanshinone, an orally bioactive herbal compound from Danshen, attenuates atherosclerosis in apolipoprotein E‐deficient mice: role of lectin‐like oxidized LDL receptor‐1 (LOX‐1)
Investigating the protective effects of CTS on atherosclerosis and its molecular mechanisms of action found it to be a major bioactive diterpenoid isolated from Danshen, an eminent medicinal herb.
Icariin attenuates the enhanced prothrombotic state in atherosclerotic rabbits independently of its lipid-lowering effects.
Icariin has lipid-lowering effects and may be used in the treatment and prevention of thrombosis in the atherosclerotic process and improves unfavorable hemorheological parameters in hypercholesterolemia.
Immunoregulatory effects of the herba Epimediia glycoside icariin.
Findings provide evidence that ICA could be applied to adoptive immunotherapy, and generation of LAK cells in presence of an appropriate dose of ICA might be superior to interleukin-2 alone.
Icariin, a natural flavonol glycoside, extends healthspan in mice
Icariin Attenuates High-cholesterol Diet Induced Atherosclerosis in Rats by Inhibition of Inflammatory Response and p38 MAPK Signaling Pathway
Icariin inhibited the HCD-induced dyslipidemia in rats, the mechanisms may be associated with the anti-inflammation, anti-oxidative stress, and downregulation of p-p38 MAPK by icariin.
Anti‐ageing active ingredients from herbs and nutraceuticals used in traditional Chinese medicine: pharmacological mechanisms and implications for drug discovery
The TCMs with anti‐ageing function are classified according to their action pathways, including the sirtuins, the mammalian target of rapamycin, AMP‐activated kinase and insulin/insulin‐like growth factor‐1 signalling pathway, free radicals scavenging and the resistance to DNA damage.
