A conditionally replicative adenoviral (CRAd) vector, designated as CRAd.pEgr-1-Smac, that promotes the overexpression of second mitochondria-derived activator of caspase (Smac) when stimulated by hypoxia and radiation was constructed. MDA-MB-231 cells were transfected with CRAd.pEgr-1-Smac and treated with 4-Gy X-rays. The hypoxic status in cancer cells was mimicked with the chemical reagent CoCl2. Smac protein expression was measured by a western blotting assay and cell proliferation was… Expand

Melittin induces human gastric cancer cell apoptosis via activation of mitochondrial pathway.
Melittin can induce apoptosis of human gastric cancer cells through the mitochondria pathways, and it may be a potent agent in the treatment of human GC. Expand
Indomethacin induces apoptosis in the EC109 esophageal cancer cell line by releasing second mitochondria-derived activator of caspase and activating caspase-3.
It is indicated that Smac is essential for the apoptosis induced by indomethacin in esophageal cancer cells, as a significant inhibition in cell viability was observed in the EC109 cells following treatment with different concentrations of indometHacin. Expand

Conditionally replicating adenovirus combined with gene-targeted radiotherapy induces apoptosis via TRAIL death receptors in MDA-MB-231 cells.
Investigation of the effects of conditionally replicating adenovirus (CRAd) in combination with X-ray irradiation on the proliferation and apoptosis of MDA-MB-231 cells showed that CRAd, in combinationWith irradiation, inhibited cell proliferation, promoted cell apoptosis and significantly increased the expression of TRAIL, DR5, caspase-3 and caspases-8 mRNA and proteins in MDA. A Hypoxia- and α-Fetoprotein–Dependent Oncolytic Adenovirus Exhibits Specific Killing of Hepatocellular Carcinomas
The results presented here show the advantages of incorporating HREs into a hAFP promoter–driven oncolytic virus and greatly enhanced selectivity and tumoricidal activity of Ad-HRE12/hAFPΔ19 make it a promising therapeutic agent in the treatment of liver cancers. Expand
[HSV-TK gene therapy of lung adenocarcinoma xenografts using a hypoxia/radiation dual-sensitive promoter].
The hypoxia/radiation dual-sensitive promoter HRE-Egr can enhance the HSV-TK gene expression in solid tumors under hypoxIA and improve the efficacy of radiogenetic therapy for lung cancer. Expand
Smac agonists sensitize for Apo2L/TRAIL- or anticancer drug-induced apoptosis and induce regression of malignant glioma in vivo
Smac agonists are promising candidates for cancer therapy by potentiating cytotoxic therapies by enhancing the antitumor activity of Apo-2L/tumor necrosis factor–related apoptosis-inducing ligand (TRAIL) in an intracranial malignant glioma xenograft model in vivo. Expand

Conditionally Replicating Adenovirus Expressing TIMP2 Increases Survival in a Mouse Model of Disseminated Ovarian Cancer
Results of the in vivo study support the translational potential of Ad5/3-CXCR4-TIMP2 for treatment of human patients with advanced ovarian cancer and confirm the therapeutic effect was confirmed to be mediated through inhibition of MMP9. Expand
Effect of pEgr-TNFalpha gene radiotherapy on mice melanoma.
It is concluded that pEgr-TNFalpha gene radiotherapy may significantly inhibit tumour growth, and that the anti-melanoma effect is superior to that of either gene therapy or radiotherapy alone. Expand
