SummaryL-5-Hydroxytryptophan (5HTP) and its major metabolites 5-hydroxytryptamine (5HT) and 5-hydroxyindoleaceticacid (5HIAA) were measured in blood and cerebrospinal fluid from neurological patients receiving steady state treatment with 5HTP. There was accumulation of 5HT in blood platelets and 5HIAA in plasma in all patients, despite concomitant administration of the L-aromatic amino acid decarboxylase inhibitors, carbidopa and benserazide. There was no correlation between the 5HTP dose and… Expand
Serotonin a la carte: supplementation with the serotonin precursor 5-hydroxytryptophan.
TLDR
Preclinical and clinical evidence for the efficacy of 5-HTP for depression is reviewed, with emphasis on double-blind, placebo-controlled (DB-PC) trials, and safety issues with 5- HTP are reviewed,With emphasis on eosinophilia myalgia syndrome (EMS) and serotonin syndrome. Expand
Slow-release delivery enhances the pharmacological properties of oral 5-hydroxytryptophan: mouse proof-of-concept
TLDR
Preclinical data suggest that 5-HTP SR could represent a new therapeutic approach to the plethora of CNS disorders potentially treatable with a pro-serotonergic drug, and might in particular be therapeutically relevant when brain 5- HT deficiency is pathogenic and as an adjunctive augmentation therapy to SSRI therapy. Expand
Efficacy and safety of 5-Hydroxytryptophan on levodopa-induced motor complications in Parkinson's disease: A preliminary finding
TLDR
Preliminary evidence of clinical benefit of 5-HTP against LIDs in PD is provided, and larger studies with a longer treatment duration and a wider range of doses are warranted to corroborate these findings. Expand
Serotonin (5-hydroxytryptamine) in medullary thyroid carcinoma with or without pheochromocytoma.
  • F. Engbaek
  • Medicine
  • European journal of cancer & clinical oncology
  • 1985
TLDR
The results show that the MTC made only little if any contribution to the overall synthesis of serotonin in the 28 patients studied; and serotonin was produced in pheochromocytoma tissue from four patients with MEN-II syndromes with phechromocyToma. Expand
The effects of 5-hydroxytryptophan on attention and central serotonin neurochemistry in the rhesus macaque
TLDR
B baseline central serotonergic functioning may underlie and predict variation in serotonin’s effects on cognitive operation, which may help inform serotonin's role in psychopathology and help clinicians predict howserotonergic interventions will influence pathologies. Expand
Enhancement of imipramine-induced rat brain β-adrenoreceptor desensitization by subacute co-administration of trazodone, zimelidine, quipazine or 5-hydroxytryptophan
TLDR
It can be concluded that noradrenaline and serotonin are both required for the process of the desensitization of central β-adrenoceptor systems by antidepressants. Expand
Interaction of 5-hydroxy-l-tryptophan and negative dietary cation-anion difference on calcium homeostasis in multiparous peripartum dairy cows.
TLDR
It is demonstrated that feeding a -DCAD diet in conjunction with 5-HTP prepartum can increase postpartum circulating iCa concentrations and therefore diminish the magnitude of hypocalcemia at the time of parturition. Expand
TREATING OBESITY AND FOOD CRAVING
increase brain serotonin (5-HT) are effective anorectic agents which help obese patients lose weight and which also decrease craving for Sweets and carbohydrates. Evidence from other studies alsoExpand
Improved strength on 5-hydroxytryptophan and carbidopa in spinal cord atrophy
TLDR
Two sisters with hemiplegic migraine, low CSF and platelet serotonin levels, and progressive spastic paraparesis associated with profound spinal cord atrophy whose lower extremity strength and ambulation responded to a precursor replacement strategy (5-hydroxytryptophan and carbidopa administration) are described. Expand
The role of human spliceosomal component SRm300 in neurodegenerative disease, response to serotonin treatment.
TLDR
This research project started in the clinic, with a family with an apparently genetic neurodegenerative disorder, including hemiplegic migraines and systemic serotonin deficiency, and found treatment with serotonin replacement significantly improved the lower limb strength. Expand

Plasma accumulation of metabolism of orally administered single dose L-5-hydroxytryptophan in man.
TLDR
The ratio of 5-HTP to carbidopa influenced the systemic bioavailability of single dose administered 5-hydroxytryptophan indicating dose dependent absorption kinetics, and co-administration of L-dopa with 5- HTP and decarboxylase inhibitors had no effect on gastrointestinal absorption in six parkinsonian patients. Expand
The effects of aromatic amino acid decarboxylase inhibitors on plasma concentrations of 5-hydroxytryptophan in man.
5-Hydroxytryptophan has been found to be beneficial when administered alone or in combination with extracerebral aromatic amino acid decarboxylase inhibitors for therapeutic purposes in variousExpand
Bioavailability and related pharmacokinetics in man of orally administered L-5-hydroxytryptophan in steady state.
TLDR
The finding of a direct proportionality between the size of the oral dose level of L-5-hydroxytryptophan and the corresponding areas under the plasma concentration curves within a dosage interval at steady state strongly indicates dose independent, linear pharmacokinetics of the compound. Expand
Plasma protein binding of tricyclic anti-depressants in man.
TLDR
The degree of binding over the entire range of therapeutic plasma drug concentration was relatively constant and the acetyl derivatives of DMI, NT and PT were much more bound than the parent compounds. Expand
Intravenous L-5-hydroxytryptophan in normal subjects: an interdisciplinary precursor loading study. Part III: Neuroendocrinological and biochemical changes.
In a precursor study with i.v. 1-5HTP (Ro 3-5940) after peripheral decarboxylase inhibition with benserazide (Ro 4-4602) in healthy human subjects the following biochemical changes were found: 1.Expand
Serotonin and myoclonus.
TLDR
It is suggested that a deficiency of brain serotonin is causally related to myoclonic muscle movements and the therapeutic efficacy of L-5-HTP plus carbidopa may be due to the repletion of serotonin in regions of the brain where serotoninergic neurons have degenerated. Expand
5-Hydroxytryptophan decarboxylase: preparation and properties.
TLDR
The studies reported in this paper concern the enzyme, 5-hydroxytryptophan decarboxylase, and procedures for purification of the enzyme obtained from hog and guinea pig kidney homogenates and acetone-dried powders and some properties of these preparations are described. Expand

TLDR
In vivo, the seryl derivative causes in vivo no or only weak inhibition of other enzymes, such as monoamine oxidase, diamine oxid enzyme, catecholamine-O-methyl transferase, transaminase of aromatic amino acids, tryptophan hydroxylase; in vitro, the Seryl derivative does not affect the histidine decarboxylase or dopamine-β-oxidase. Mechanism of intestinal absorption and brain uptake of L-5-hydroxytryptophan in rats, as compared to those of L-3,4-dihydroxyphenylalanine.
TLDR
In vitro studies using rat brain cortex slices proved that both L-5-HTP and L-DOPA, but not their D-isomers, are taken up by the brain tissues by an active transport mechanism which is saturable, energy-dependent and competitive. Expand
