The functions involved in strain-specific modification and restriction of DNA produced by Escherichia coli are under the genetic control of the chromosome or of other genetic elements such as prophage and transfer factors. ' They are active upon bacterial as well as many phage DNA's. The relatively small, biologically active phage DNA's which can be isolated in a homogeneous form provide a convenient system for studying the molecular mechanism of the functions.
