Primary hamster embryo cells were grown in vitro with Dulbecco's minimal essential medium supplemented with 10% dialyzed, virus-screened fetal calf serum and adjusted to 0.1 mm Ca++. Cultures were segregated according to sex. Human adenovirus type 12 (Ad-12) induced foci of morphologically transformed cells in both sexes 10 to 12 days postinfection. The number of foci in replicate cultures did not vary statistically. Consistently more foci developed in female cultures than in similarly infected…

The results suggest that following adenovirus transformation there is a spontaneous progression in the expression of markers of transformation and that the phorbol ester tumor promoters can accelerate this process.

It was shown that FeLV-FeSV can induce morphological transformation in human cells in vitro and that there is a requirement for the cells to passage through soft agar before subsequent tumor formation (neoplastic transformation) can be demonstrated.

Fetal bovine sera from each of three different commercial sources were tested for their ability to support cloning of human fibroblastoid cells in vitro and addition of estrogen or hydrocortisone to a culture medium that inhibits growth, with subsequent reversal of the inhibitory effect, implies that these hormones competitively regulate growth of responsive cells in vivo.

Evidence is suggestive of a posttranscriptional effect possibly modulating viral genome expression resulting in an increased efficiency of viral transformation, and an increased proliferation of transformed cells.

Hormonal reversal of this resistance resulted in enzyme profiles that reflected the pattern of young susceptible cells, and changes in enzyme patterns indicated basic alterations in protein synthesis.

With the exception of ZnCl2 and ZnSO4, enhancement was demonstrated by both a relative increase in the viral transformation frequency and an absolute increase inThe number of transformed foci among treated cells negate the possibility that enhancement resulted from the selection of transformation-sensitive cells.
