Although plasminogen activator inhibitor 1 (PAI-1) is one of the primary regulators of the fibrinolytic system, it also has dramatic effects on cell adhesion, detachment and migration. PAI-1 also differs from other serine protease inhibitors (serpins) in that it is a trace protein in plasma, it has a short half-life in vivo, its synthesis is highly regulated, and it binds to the adhesive glycoprotein vitronectin (VN) with high affinity and specificity. These unique and diverse properties of PAI…
Small Molecule Targeting of PAI-1 Function: A New Therapeutic Approach for Treatment of Vascular Stenosis
TLDR
These findings implicate PAI-1 as a significant factor in the development of restenosis and provided the impetus for development of low-molecular weight PAi-1 antagonists.
Three decades of research on plasminogen activator inhibitor-1: a multifaceted serpin.
TLDR
The review provides an overview on the current knowledge on PAI-1, a unique member of the serpin superfamily, exhibiting particular unique conformational and functional properties in the plasminogen-plasmin system.
Resolving distinct molecular origins for copper effects on PAI-1
TLDR
A gel-based copper sensitivity assay is developed that can be used to assess the copper concentrations that accelerate the conversion of active PAI-1 to a latent form, and identified a copper-binding site involving histidines at positions 2 and 3 that confers a remarkable stabilization of PAi-1 beyond what is observed with vitronectin alone.
Biochemical properties of plasminogen activator inhibitor-1.
TLDR
Studies of PAI-1 have contributed significantly to the elucidation of the protease inhibitory mechanism of serpins, which is based on a metastable native state becoming stabilised by insertion of the RCL into the central beta-sheet A and formation of covalent complexes with target proteases.
Emerging role of serpinE2/protease nexin-1 in hemostasis and vascular biology.
TLDR
Recent data underlining the emerging role of PN-1 as a key factor in the responses of vessels to injury are reviewed, providing insights that may allow the development of drugs and treatment strategies to prevent or limit thrombotic disorders.
Plasminogen Activator Inhibitor 1 in Platelets - Studies of Synthesis, Activity, and Glycosylation Patterns
TLDR
The role of platelets in inhibition of fibrinolysis, and in particular the physiological regulation of platelet-derived PAI-1; its synthesis, activity, and potential contribution to plasma levels is investigated to clarify the previous irreconcilable findings.
Plasminogen activator inhibitor-1 in cancer research.
Inhibition of Plasminogen Activator Inhibitor-1: Its Mechanism and Effectiveness on Coagulation and Fibrosis
TLDR
Beneficial in vitro and in vivo effects of novel PAI-1 inhibitors are demonstrated and the methodology proves to be a useful tool to obtain effective inhibitors of serpin activity.
Plasminogen activator inhibitor-1 is locked in active conformation and polymerizes upon binding ligands neutralizing its activity.
TLDR
It is found that some inhibitors of PAI-1 arrest this serpin in its active form instead of increasing the speed of conversion, which abolishing a necessary step in the conversion of this protein into the latent form.

The structural basis for the pathophysiological relevance of PAI-I in cardiovascular diseases and the development of potential PAI-I inhibitors.
TLDR
An overview of the knowledge on PAI-I gathered since its discovery two decades ago is provided, discussing its structural properties of the protein and their subsequent relation to functional activities, and its role in a wide variety of (patho)physiological processes.
Biological and clinical aspects of plasminogen activator inhibitor type 2.
TLDR
The aim of the present review is to bring together recent findings concerning the properties of PAL2, its gene structure, its cellular distribution, the regulation of its gene expression, and its potential role in pregnancy, skin, inflammation, wound healing, tumor invasion, and metastasis.
Plasminogen activator inhibitor-1 is a major stress-regulated gene: Implications for stress-induced thrombosis in aged individuals
TLDR
The results suggest that the large induction of PAI-1 by stress increases the risk for thrombosis in the older populations, and that the adipose tissue may be involved.
Plasminogen Activator Inhibitor-1 and -3 Increase Cell Adhesion and Motility of MDA-MB-435 Breast Cancer Cells*
TLDR
It is reported herein that PAI-1, when added exogenously to or stably transfected in human MDA-MB-435 breast carcinoma cells, had disparate effects on adhesion to extracellular matrix proteins and motility in vitro and introducedPAI-3 as another serpin potentially important in malignant disease.
Plasminogen activator inhibitor-1 in tumor growth, angiogenesis and vascular remodeling.
TLDR
PAI-1 appears to have a multifunctional role in regulating the migratory and fibrinolytic activity of vascular cells, and this, in turn, may help to explain the many varied actions of PAi-1.
Distribution of plasminogen activator inhibitor (PAI-1) in tissues.
TLDR
PAI-1 occurred throughout the body; its concentration and activity differed considerably from organ to organ, but the activity of the inhibitor was much higher in liver than in spleen: the liver may be a source of plasma PAI- 1.
The adipocyte and hemostatic balance in obesity: studies of PAI-1.
TLDR
Preliminary studies indicate that TF gene expression is elevated in the adipose tissues of the obese mouse, and this raises the possibility that the abnormal expression of other hemostatic genes by adipocytes in obesity/NIDDM may also contribute to the cardiovascular complications of this disorder.
The Low Density Lipoprotein Receptor-related Protein Is a Motogenic Receptor for Plasminogen Activator Inhibitor-1*
TLDR
Results indicate that LRP is a signaling molecule, that it mediates the migration-promoting activity of PAi-1, and that this activity does not require intact, biologically active PAI-1.
Plasminogen activator inhibitor-1 and vitronectin promote vascular thrombosis in mice.
TLDR
The results suggest that endogenous fibrinolysis and its regulation by PAI-1 and VN have important roles in the development of occlusive vascular thrombosis after vascular injury.
