Signaling pathways transmit information through protein interaction networks that are dynamically regulated by complex extracellular cues. We developed LUMIER (for luminescence-based mammalian interactome mapping), an automated high-throughput technology, to map protein-protein interaction networks systematically in mammalian cells and applied it to the transforming growth factor–β (TGFβ) pathway. Analysis using self-organizing maps and k-means clustering identified links of the TGFβ pathway to…

Regulation of the Polarity Protein Par6 by TGFß Receptors Controls Epithelial Cell Plasticity
- BiologyScience
- 2005
It is demonstrated that Par6, a regulator of epithelial cell polarity and tight-junction assembly, interacts with TGFβ receptors and is a substrate of the type II receptor, TβRII, which controls the interaction of Par6 with the E3 ubiquitin ligase Smurf1.

Small interfering RNA-targeting aPKC reduces TGF-β-induced RhoA and E-cadherin loss, cell morphology changes, stress fiber production, and the migration of NSCLC cells, and suggests that aPKCs cooperate with T GF-β receptors to regulate phospho-Par6-dependent EMT and cell migration.
