This study was undertaken to examine the regulation of leptin gene (LEP) transcription and leptin release by hexosamines in 3T3-L1 adipocytes. Glucosamine (1 mM), an intermediate in hexosamine biosynthesis, increased leptin release to 117.0 +/- 7.3% (P = 0.0430; n = 9) and 134.6 +/- 6.5% of the control value (P = 0.0367; n = 4) by 48 and 96 h, respectively. With 0.01 mM glucosamine, leptin release was increased to 120.0 +/- 3.0% of the control value (P = 0.0069; n = 4) by 96 h of treatment… Expand
Lipoic acid inhibits leptin secretion and Sp1 activity in adipocytes.
TLDR
Data suggest that the phosphorylation of Sp1 and the accompanying reduced DNA-binding activity are likely to be involved in the inhibition of leptin induced by LA, which could be mediated in part by the abrogation of the PI3K/Akt pathway. Expand
Glucosamine induces rapid desensitization of glucose transport in isolated adipocytes by increasing GlcN-6-P levels.
TLDR
It is concluded that glucosamine elevates intracellular GlcN-6-P levels within minutes, resulting in desensitization of the glucose transport system through allosteric inhibition of hexokinase, and prolonged treatment elevates Glcn- 6-P to levels that cannot be effectively lowered by cell washing. Expand
Hexosamine flux, the O-GlcNAc modification, and the development of insulin resistance in adipocytes
TLDR
Dynamic and inducible O-GlcNAc modification is emerging as a modulator of insulin sensitivity in adipocytes as a major downstream event consuming and incorporating the final product of the hexosamine biosynthesis pathway. Expand
Glucose-containing peritoneal dialysis fluids regulate leptin secretion from 3T3-L1 adipocytes.
TLDR
The results suggest that the PD-induced hyperleptinaemia could, in part, be mediated by the effect of glucose-based dialysis fluids on leptin production by adipocytes via activation of the hexosamine biosynthetic pathway. Expand
Glucosamine induces lipid accumulation and adipogenic change in C2C12 myoblasts.
TLDR
The content of uridine-5'-diphospho-N-acetylglucosamine, the end product of the HBP, was elevated in skeletal muscle of obese diabetic KKA(y) mice, compared with control mice, providing a possible explanation of hyperglycemia-induced insulin resistance in skeletal muscles. Expand
Hexosamine biosynthesis impairs insulin action via a cholesterolgenic response.
TLDR
Data suggest a novel mechanism whereby increased HBP activity increases Sp1 transcriptional activation of a cholesterolgenic program, thereby elevating plasma membrane cholesterol and compromising cytoskeletal structure essential for insulin action. Expand
Global O-GlcNAc Levels Modulate Transcription of the Adipocyte Secretome during Chronic Insulin Resistance
TLDR
It is shown that upon the elevation of O-GlcNAc levels and the induction of IR in mature 3T3-F442a adipocytes, the transcript levels of multiple secreted proteins reflect the modulation observed at the protein level, and this work confirms that Sp1 is a common cis-acting element. Expand
Chito-Oligosaccharide Inhibits the De-Methylation of a ‘CpG’ Island within the Leptin (LEP) Promoter during Adipogenesis of 3T3-L1 Cells
TLDR
It is suggested that COS can have an epigenetic effect on differentiating adipocytes, a novel biological function of COS which has potential applications for the manipulation of leptin gene expression, adipogenesis, and conditions within the metabolic syndrome spectrum. Expand
Sp1-mediated transcription is involved in the induction of leptin by insulin-stimulated glucose metabolism.
TLDR
A key role for Sp1 is supported in the transcriptional activation of the leptin gene promoter by insulin-mediated glucose metabolism, as mediated by a previously identified Sp1 site. Expand
Links between de novo fatty acid synthesis and leptin secretion in bovine adipocytes.
TLDR
Examination of the effect of cerulenin, an inhibitor of de novo fatty acid synthesis at the step from malonyl CoA to palmitate, on leptin production by cultured bovine adipocytes derived from intermuscular fat concludes that, as in monogastrics, malonyL CoA may play a key role in the control of leptin secretion in ruminants. Transcriptional regulation of the leptin promoter by insulin-stimulated glucose metabolism in 3t3-l1 adipocytes.

Glucosamine-induced Insulin Resistance in 3T3-L1 Adipocytes Is Caused by Depletion of Intracellular ATP*
TLDR
Direct administration of glucosamine can rapidly lower cellular ATP levels and affect insulin action in fat cells by mechanisms independent of increased intracellular UDP-N-acetylhexosamines and that increased metabolism of glucose via the hexosamine pathway may not represent the mechanism of glucose toxicity infat cells.
