Vascular endothelial growth factor (VEGF) inhibits radiation-induced apoptosis in the leukemia cell line, CMK86 (O. Katohet al.,Cancer Res., 55: 5687–5692, 1995). To elucidate the molecular mechanisms underlying this inhibitory effect of VEGF, we attempted to identify a transcription factor involved in the cellular response to VEGF stimulation. We cloned the cDNA of a novel Krüppel-type zinc finger (ZNF) gene,ZK7, from a cDNA library constructed from the human leukemia cell line CMK86 after… Expand

De novo microduplication of the FMR1 gene in a patient with developmental delay, epilepsy and hyperactivity
The case of a child who presented with developmental delay starting at 9 months of age, fine motor and speech delay, progressive seizures since 18 years of age and hyperactivity is reported, including a de novo microduplication in the Xq27.3 region, including the FMR1 gene and the ASF MR1 gene. ZK1, a novel Krüppel-type zinc finger gene, is induced following exposure to ionizing radiation and enhances apoptotic cell death on hematopoietic cells.
The cloned cDNA of a novel Krüppel-type zinc finger (ZNF) gene, ZK1, from a cDNA library constructed from the human leukemia cell line, CMK86, using degenerate primers suggests that this gene is one of early response genes by exposure to ionizing radiation and may have some functions on radiation-induced apoptotic cell death on hematopoietic cells. Vascular endothelial growth factor inhibits apoptotic death in hematopoietic cells after exposure to chemotherapeutic drugs by inducing MCL1 acting as an antiapoptotic factor.
It is shown that VEGF also inhibits apoptotic cell death that is induced by exposure to the chemotherapeutic drugs etoposide and doxorubicin and the expression level of MCL1, a member of the BCL2 family, was increased by V EGF. Expand
Expression of vascular endothelial growth factor and its receptors in hematopoietic malignancies.
The results raise the possibility that VEGF may play a role in the growth of hematopoietic neoplasms such as multiple myeloma through either a paracrine or an autocrine mechanism. Expand
Expression of the vascular endothelial growth factor (VEGF) receptor gene, KDR, in hematopoietic cells and inhibitory effect of VEGF on apoptotic cell death caused by ionizing radiation.
The expression of the KDR gene transcript is demonstrated, which encodes a cell surface receptor for VEGF, in normal human hematopoietic stem cells, megakaryocytes, and platelets as well as in human leukemia cell lines, HEL and CMK86, suggesting that V EGF may give leukemia cells some abilities of resistance against radiotherapy in an autocrine or paracrine manner. Overexpression of the zinc finger protein MZF1 inhibits hematopoietic development from embryonic stem cells: correlation with negative regulation of CD34 and c-myb promoter activity
R reverse transcription-PCR analysis of human peripheral blood CD34+ cells cultured under lineage-restricted conditions demonstrated MZF1 expression during both myeloid and erythroid differentiation, indicating that MZf1 plays a critical role in hematopoiesis by modulating the expression of genes involved in this process. Up-regulation of vascular endothelial growth factor/vascular permeability factor in mouse skin carcinogenesis correlates with malignant progression state and activated H-ras expression levels.
Data show that in mouse skin carcinogenesis, the VEGF/VPF angiogenic stimulus occurs early during premalignant papilloma development and further increases at later stages, and it is demonstrated that increasing the activated H-ras dose may play an additional role by facilitating malignant in vivo progression through the modulation of V EGF/ VPF-mediated angiogenesis. Vascular permeability factor/vascular endothelial growth factor-mediated signaling in mouse mesentery vascular endothelium.
A new model system with the potential for investigating signaling events in response to diverse mediators on intact microvessels in vivo is described and further elucidated the VPF/VEGF signaling cascade. Expand
The Krüppel-type zinc finger family gene, HKR1, is induced in lung cancer by exposure to platinum drugs.
It is found that mRNA expression of the HKR1 gene was induced in lung-cancer cell lines by exposure to cisplatin using Northern blot analysis, and that high expression levels in lung cancers were associated with antemortem platinum drug administration. Expand
A novel zinc finger gene on human chromosome 1qter that is alternatively spliced in human tissues and cell lines.
Two novel zinc finger-encoding cDNAs from a human hepatoblastoma cell line are isolated and characterized, which demonstrate DNA sequence homology to a recently described human Kruppel-related gene HZF-3 and appear to be derived from a single gene by alternate mRNA splicing. Expand
Identification and characterization of the Egr-1 gene product, a DNA-binding zinc finger protein induced by differentiation and growth signals.
It is shown by cell fractionation and immunocytochemistry that the Egr-1 protein is located in the nucleus and that it is phosphorylated, and in vitro-generated Egr -1 protein binds with high affinity to the sequence CGCCCCCGC in a zinc-dependent manner. Expand
